Published: April 2023

Seven new 18-residue peptaibols, trichorzins A-G (-), were isolated from the sponge-derived fungus sp. GXIMD 01001. Their structures and configurations were characterized by a comprehensive interpretation of the NMR spectroscopic data, MS/MS fragmentation, Marfey's method, and ECD analysis. The general sequences of trichorzins A-G are as follows: Ac-Aib-Ala/Ser-Ala-Aib/Iva-Iva-Gln-Aib/Iva-Val/-Ile-Aib-Gly-Leu-Aib-Pro-Leu-Aib-Aib-Gln-Trpol/Pheol. The obtained compounds were assessed for their potential antiproliferative and antimicrobial activities. All obtained compounds did not show potent antibacterial activity but exhibited significant cytotoxicity, with the lowest IC values at 0.46-4.7 μM against four human carcinoma cell lines.

Trichorzin PA is a family of 18-residue peptaibols with linear and flexible peptide chains. The three-dimensional structures and biological activities of trichorzin PA peptaibols are largely uncharacterised. In this work, two previously identified peptaibols, trichorzin PA VI () and II (), were isolated from ML-P8-2.

We previously reported on a novel peptaibol, named Tripleurin XIIc (TPN), an 18-residue long sequence produced by the fungus . We elucidated its 3D structure via classical and accelerated molecular dynamics simulation (aMD) methods and reported the folding dynamics of TPN in water and chloroform solvents. Peptaibols, in general, are insoluble in water, as they are amphipathic and may prefer hydrophobic environments like transmembrane regions.

Article Synopsis
  • * The structural configurations for the new peptaibols were determined using techniques like nuclear magnetic resonance (NMR) and high-resolution mass spectrometry, while their absolute configurations were assessed using Marfey's method.
  • * All 18-residue peptaibols showed moderate antibacterial activity against Staphylococcus aureus, with MIC values between 8-32 μg·mL, while one 14-res

Buildings that have been flooded often have high concentrations of Trichoderma spores in the air while drying. Inhaled spores and spore and mycelial fragments contain large amounts of fungal glucan and natural products that contribute to the symptoms associated with indoor mould exposures. In this study, we considered both small molecules and peptaibol profiles of T.
