We report a case of a 49 year old woman who developed biopsy-proven xanthomas on her hands and arms after initiation of Mycophenolate mofetil therapy for systemic lupus erythematosus (SLE) and subsequently went into remission upon cessation of the medication.

Previous attempts to identify neuroprotective targets for acute ischemic stroke by studying ischemic cascades and devising ways to suppress these pathways have failed in translational research. We hypothesized that studying the molecular determinants of endogenous neuroprotection, namely, the tolerance against ischemic stroke conferred by hyperbaric oxygen (HBO) preconditioning, via a well-established paradigm would reveal new neuroprotective targets. By a combination of proteomics, KEGG pathway analysis, lysosome fraction and western blot analysis, we found that chaperone-mediated autophagy (CMA) was activated by HBO preconditioning.

Article Synopsis
  • Mycophenolate mofetil (MMF) is commonly used as an immunosuppressant in transplantation but can cause gastrointestinal toxicity, with limited understanding of its metabolic effects.
  • In this study, mice were treated with MMF for two weeks, and a variety of metabolic changes were analyzed using gas chromatography-mass spectrometry and specific databases to understand the impacts on various organs.
  • The results revealed that MMF treatment altered metabolite levels in several organs and disrupted amino acid and fatty acid metabolism, while also increasing certain immune cell types without significantly affecting others.

Article Synopsis
  • Mycophenolate mofetil (MMF) is a common immunosuppressive drug used in organ transplants but causes gastrointestinal (GI) side effects that are not well understood.
  • Research indicates that gut microbiota and their metabolites, particularly secondary bile acids (BAs), are significantly affected by MMF, which is linked to GI toxicity.
  • The study highlights that supplementing with lithocholic acid can reduce MMF's negative effects on GI health, and the use of paricalcitol, a VDR agonist, may provide a potential treatment option for these side effects.
