A divergent approach to a small library of long-chain 6-amino-1,4,5-triols as novel phytosphingosine-type entities, together with their preliminary cytotoxic evaluation, was achieved. Construction of the target compounds addressed two key aspects. First, the installation of a carbon-nitrogen bond via two prototypes of [3,3]-sigmatropic rearrangements and second the introduction of an alkyl side chain unit by using a late stage olefin cross-metathesis process. As shown in cell viability experiments, the corresponding HCl salts proved to be the most cytotoxic derivatives among all the tested substances, with IC values in the lower micromolar range on the Jurkat, HeLa and HCT-116 cell lines.

In this work, we present an efficient strategy for the straightforward synthesis of functionalized 1,6-dihydropyridine derivatives a three-component reaction of 3-vinylchromones, aromatic aldehydes, and ammonium acetate. A tandem procedure including NH aldimine formation/Michael-type addition/opening of the pyrone ring/isomerization/6π-electrocyclization/[1,5]-H shift allows rapid access to a series of dihydropyridines bearing an -hydroxybenzoyl and a benzoyl scaffold in good yields. Readily available precursors, simple heating conditions, and operational simplicity are some highlighted advantages of this transformation.

The Ph3P-I2-mediated reactions between isatins and amines were extensively investigated leading to the discovery of highly selective and divergent routes toward the synthesis of two distinct classes of indole-based frameworks. Through a strategic design of the reaction paths, we overcome potential side reactions to achieve convenient and straightforward one-pot methods to access either indoloquinazolines with C-12 carboxamide or 2-aminosubstituted indol-3-ones using the same reagent system. Mechanistic studies reveal the role of Ph3P-I2 in governing product selectivity, providing an efficient route to novel fused-indolone derivatives with promising applications in drug discovery and medicinal chemistry.

A palladium-catalyzed alkoxycarbonylation with two different alcohols for the synthesis of cyclobutanecarboxylates bearing an α-quaternary carbon center is presented. The reaction utilizes readily accessible starting materials, tolerates a broad scope of functional groups, and provides a straightforward and efficient approach for the synthesis of a diverse array of cyclobutanecarboxylates bearing an α-quaternary carbon. Meanwhile, this strategy effectively prevents the transition-metal-catalyzed ring-opening of cyclobutanols, preserves the cyclobutane framework, and affords 1,1-disubstituted cyclobutanecarboxylates in high yields with excellent regioisomeric ratios.
