The vacuole is one of the most conspicuous organelles in plant cells, participating in a series of physiological processes, such as storage of ions and compartmentalization of heavy metals. Isolation of intact vacuoles and elemental analysis provides a powerful method to investigate the functions and regulatory mechanisms of tonoplast transporters. Here, we present a protocol to isolate intact vacuoles from root protoplasts and analyze their elemental content by inductively coupled plasma mass spectrometry (ICP-MS). In this protocol, we summarize how to prepare the protoplast, extract the vacuole, and analyze element concentration. This protocol has been applied to explore the function and regulatory mechanisms of tonoplast manganese (Mn) transporter MTP8, which is antagonistically regulated by CPK4/5/6/11 and CBL2/3-CIPK3/9/26. This protocol is not only suitable for exploring the functions and regulatory mechanisms of tonoplast transporters, but also for researching other tonoplast proteins. Graphical abstract.

Many cells spend a major part of their life in quiescence, a reversible state characterized by a distinct cellular organization and metabolism. In glucose-depleted quiescent yeast cells, there is a metabolic shift from glycolysis to mitochondrial respiration, and a large fraction of proteasomes are reorganized into cytoplasmic granules containing disassembled particles. Given these changes, the operation of protein quality control (PQC) in quiescent cells, in particular the reliance on degradation-mediated PQC and the specific pathways involved, remains unclear.

Article Synopsis
  • Pathogenic mutations in genes associated with tuberous sclerosis complex (TSC) affect various types of kidney tumors like eosinophilic solid and cystic renal cell carcinoma (ESC RCC) and oncocytic tumors.
  • A study validated a TSC2 immunohistochemistry (IHC) assay on tissues from renal tumors to assess the presence of TSC2 mutations, achieving high predictive values for diagnosing these tumors.
  • The findings highlight that TSC2 IHC can help identify renal tumors linked to TSC/mTOR pathway mutations, supporting its use in diagnostic processes.

Our study seeks to provide a theoretical foundation for the clinical use of cold-stored platelets (CSPs) by interpreting ultrastructural images and quantitatively analyzing structural changes. CSPs, room temperature-stored platelets (RTPs), and delayed CSPs (delayed-CSPs) were continuously observed using scanning electron microscopy and transmission electron microscopy at eight time points. Super-resolution fluorescence microscopy was employed to observe changes in platelet microtubules and mitochondrial structure and function, whereas platelet counts, metabolism, and relevant functional indicators were measured concurrently.

Article Synopsis
  • - The study aimed to investigate how electroacupuncture (EA) affects the HIF/VEGF/Notch signaling pathways in rats with cerebral ischemia, focusing on its potential role in enhancing cerebral microangiogenesis and improving brain function.
  • - It included 100 male SD rats, with 15 in a sham group and the remaining 85 subjected to a cerebral ischemia model; various therapies, including EA and specific inhibitors, were administered to study their effects on neurological deficits and brain tissue characteristics.
  • - Results showed significant differences in neurological scores, cerebral infarct volume, microvascular density, and the expression of key proteins related to the signaling pathways in rats treated with EA compared to controls, highlighting the beneficial effects of EA
