Age-related muscle wasting, sarcopenia is an extensive loss of muscle mass and strength with age and a major cause of disability and accidents in the elderly. Mechanisms purported to be involved in muscle ageing and sarcopenia are numerous but poorly understood, necessitating deeper study. Hence, we employed high-throughput RNA sequencing to survey the global changes in protein-coding gene expression occurring in skeletal muscle with age.

Modeling the lymph node stromal cells in oral squamous cell carcinoma: insights into the stromal cues in nodal metastasis.

Hum Cell

January 2025

Integrated Head and Neck Oncology Program (DSRG-5), Mazumdar Shaw Medical Foundation, Narayana Health, Bangalore, India.

Article Synopsis
  • The study investigates lymph node stromal cell cultures (LNSCs) from oral squamous cell carcinoma (OSCC) patients to understand tumor interactions for better treatment options.
  • Primary cultures were developed from both metastatic and non-metastatic patient nodes, confirming their purity and revealing a variety of cell types.
  • Transcriptomic and cytokine profiling showed different molecular alterations based on node status, with key insights into immune response and potential pathways for metastasis, highlighting the relevance of these models for future cancer research.

Self-amplifying RNA (saRNA) is an extremely promising platform because it can produce more protein for less RNA. We used a sort and sequence approach to identify host cell factors associated with transgene expression from saRNA; the hypothesis was that cells with different expression levels would have different transcriptomes. We tested this in CDK4/hTERT immortalized human muscle cells transfected with Venezuelan equine encephalitis virus (VEEV)-derived saRNA encoding GFP.

Background: Peanut allergy (PA) is one of the most prevalent food allergies with a lack of favorable safety/efficacy treatment. A cucumber mosaic virus-like particle expressing peanut allergen component Ara h 2 (VLP Peanut) has been developed as a novel therapeutic approach for PA.

Objective: We assessed the tolerogenic properties and reactivity of VLP Peanut.

Article Synopsis
  • Antiphospholipid antibody syndrome (APS) is an autoimmune disease that causes blood vessel problems and can lead to serious kidney damage known as nephropathy.
  • Researchers tested a Tyk2 inhibitor, BMS-986202, on mice with APS nephropathy to assess its effects on the disease.
  • The study found that Tyk2 inhibition not only improved kidney function and reversed harmful changes in the kidneys, but also reduced the type I interferon (IFN) signature associated with the disease, indicating a potential new treatment approach for APS nephropathy.
