Tofactinib is a JAK inhibitor approved for ulcerative colitis in humans. Despite of its' proven effectiveness in humans, mechanistic data are scarce on the effectiveness of Tofactinib in experimental colitis in mice. We induced experimental colitis by transfer of CD4+CD25- isolated T cells into RAG2-/- (T and B cell deficient) mice and treated these mice with tofacitinib for 5-6 weeks either with a dosage of 10 or 40 mg/kg body weight immediately after CD4+ transfer or started treatment after first symptoms of disease for several weeks. While treatment with tofacitinib immediately after transfer resulted in an enhanced expansion of CD4+ T cells and did not prevent occurrence of colitis, treatment after start of symptoms of colitis ameliorated disease activity on a clinical basis and in histological analyses. Tofacitinib is effective in the treatment of murine experimental T cell transfer colitis, however does not prevent occurrence of disease.

Purpose: This study aimed to identify a novel recombinant adeno-associated virus (rAAV) capsid variant that can widely transfect the deep retina through intravitreal injection and to assess their effectiveness and safety in gene delivery.
Methods: By adopting the sequences of various cell-penetrating peptides and inserting them into the capsid modification region of AAV2, we generated several novel variants. The green fluorescent protein (GFP)-carrying variants were screened following intravitreal injection.

Two new nonfused ring nonfullerene electron acceptors, NFAs, (dicarbazolyl)bis(2-(3-oxo-2,3-dihydro-1-inden-1-ylidene)malononitrile) () and -(2-(5,6-fluoro-3-oxo-2,3-dihydro-1-inden-1-ylidene)malononitrile) (), thus exhibiting an A-D-A motif, were synthesized and characterized. As thin films, they exhibit the lowest energy absorption signature near 540 nm, extending down to ∼700 nm. This band is due to an intramolecular charge transfer process from the (nonfused dicarbazoyl; ) moiety to the malononitrile-based units () based on density functional theory calculations (DFT), which are also corroborated by time-dependent DFT (TDDFT) computations.

Ternary solar cells have been rapidly developed in the realm of organic solar cells (OSCs). The incorporation of a third component into a cell results in a complicated active layer morphology, and the relation of this morphology to power conversion efficiency remains elusive. In this work, two ternary active layers, B1:Y7 (10 wt%):BO-4Cl and B1:Y7 (50 wt%):BO-4Cl are constructed, and the reasons for the differences in PCE caused by varying the Y7 content are investigated using theoretical calculations.
