The paper deals with the evaluation of quantitative indexes of urine excretion of tyrosine and its metabolites--highly carcinogenic p-hydroxyphenyllactic acid and homogentisic acid--in infants suffering various tumors and their parents. Also, the effect of ascorbic acid (100 mg/kg body weight daily) on tyrosine metabolism was studied. The sick children and their parents showed increased urine excretion of tyrosine and p-hydroxyphenyllactic acid matched by a decrease in that of homogentisic acid. The said changes suggest inhibition of p-hydroxyphenylpyruvate dioxygenase (CE 1.13.11.27). High doses of ascorbic acid were shown to arrest or significantly inhibit the excretion of carcinogenic metabolite of tyrosine in patients with hemoblastoses and nephroblastoma.

Objective: We previously found that per- and polyfluoroalkyl substances (PFAS) mixture exposure is inversely associated with SARS-CoV-2 IgG (IgG) antibody levels in pregnant individuals. Here, we aim to identify metabolites mediating this relationship to elucidate the underlying biological pathways.
Methods: We included 59 pregnant participants from a US-based pregnancy cohort.
Starvation, intermittent fasting and exercise, all of which are recommended lifestyle modifiers share a common metabolic signature, ketogenesis to generate the ketone bodies, predominantly β-hydroxybutyrate. β-hydroxybutyrate exerts beneficial effects across various contexts, preventing or mitigating disease. We hypothesized that these dynamic health benefits of β-hydroxybutyrate might stem from its ability to regulate genome architecture through chromatin remodeling via histone β-hydroxybutyrylation, thereby influencing the transcriptome.

The proliferation of CAR-T cells was hindered and cannot play its killing function well in solid tumors. And yet the regulatory mechanism of CAR-T cell proliferation is not fully understood. Here, we showed that recombinant expression of CD19CAR in T cells significantly increased the basal activation level of CAR-T cells and LCK activation.
