Loss-of-function mutations in SPOP E3 ubiquitin ligase drive multiple cancers. However, carcinogenic gain-of-function SPOP mutations have been a major puzzle. In this issue of Molecular Cell, Cuneo et al. show that several mutations map to SPOP oligomerization interfaces. Additional questions remain about SPOP mutations in malignancy.
