A new route to bicyclic γ-lactams was found, which was proposed as a three-component cyclization of ethyl trifluoropyruvate with methyl ketones and 1,2-, 1,3-amino alcohols. As a result, a series of trifluoromethyl-substituted tetrahydropyrrolo [2,1-]oxazol-5-ones and tetrahydropyrrolo[2,1-][1,3]oxazine-6-ones was synthesized, in which the substituent at the nodal carbon atom was varied. The introduction of a twofold excess of ethyl trifluoropyruvate in reactions with amino alcohols and acetone made it possible to obtain the same bicycles, but functionalized with a hydroxyester fragment, which are formed due to four-component interactions of the reagents. Transformations with 2-butanone and aminoethanol lead predominantly to similar bicycles, while an analogous reaction with aminopropanol gives -hydroxypropyl-2,3-dihydropyrrol-5-one. Almost all bicycles are formed as two diastereomers, the structure of which was determined using H, F, C NMR spectroscopy, including two-dimensional experiments and XRD analysis. A domino mechanism for the formation of tetrahydropyrrolo[2,1-]oxazacycles was proposed, which was confirmed by their stepwise synthesis through the preliminary preparation of the aldol and bis-aldol from ethyl trifluoropyruvate and methyl ketones.

A novel route for tandem C-N/C-O formation copper-catalyzed [4+2] oxidative annulation of α,β-unsaturated ketoxime acetates with ethyl trifluoropyruvate to synthesize valuable trifluoromethyl-containing 2-1,3-oxazines in moderate to good yields is developed. This procedure represents the first [4+2] oxidative annulation of oxime derivatives with activated CO bonds and provides an alternative route towards functionalized 2-1,3-oxazines.

Via difunctionalization of the α- and β-sites of cyclic ethers, we herein demonstrate a new synthetic method for the efficient construction of novel fluorinated γ-amino acid esters by employing a CoBr/-CPBA catalyst system. Several cyclic ethers were transformed in combination with a vast range of amines and ethyl trifluoropyruvate into the desired products under mild conditions, making this method a practical platform to enrich the library of fluorinated amino acid derivatives from cost-effective and readily available feedstocks.

An efficient and benign iron-catalyzed room-temperature method was developed for direct sequential C(sp)-H/N-H annulation to construct pyrroloindole scaffolds. Searching for new less toxic anticancer drug candidates is a big challenge from a medical point of view. The present investigation was aimed at describing two independent synthetic approaches based on isosteric replacements, spectroscopic characteristics, in vitro anticancer and ex vivo antihaemolytic activities of novel molecules (9-22) and correlations between their standardised lipophilicity indices, computed log P values and pharmacokinetic descriptors. Two novel protocols for annelation of the triazinone template on hydrazinylideneimidazolidines (1-8) (showing a high reactivity towards electrophilic reagents, such as ethyl trifluoropyruvate and ethyl 3-methyl-2-oxobutyrate) were developed for the first time, giving rise to two original classes of highly conjugated azaisocytosine-containing molecules (9-16 and 17-22).
