Background: The antiviral activity and underlying mechanism of Patchouli alcohol remain unclear.

Methods: This study evaluated the cytotoxicity, optimal methods for drug administration, anti-influenza A activity of Patchouli alcohol. The antiviral mechanism of Patchouli alcohol was also assessed qRT-PCR, western blot, hemagglutination inhibiting (HAI) assay, and hemolysis inhibiting assay.

Results: Patchouli alcohol was shown to have low cytotoxicity and its strongest antiviral effect was associated with premixed administration. Patchouli alcohol inhibited virus replication during the early lifecycle stages of influenza A virus infection and specifically prevented expression of the viral proteins, HA and NP. In both the HAI and hemolysis inhibiting assays, Patchouli alcohol was able to block HA2-mediated membrane fusion under low pH conditions. Patchouli alcohol had lower binding energy with HA2 than HA1.

Conclusion: These findings suggest that Patchouli alcohol could be a promising membrane fusion inhibitor for the treatment of influenza A infection.

Introduction: Prostate cancer is an androgen-dependent malignancy, and the use of androgen deprivation therapies frequently results in treatment resistance, relapse, and the development of aggressive castration-resistant tumors. Patchouli alcohol, a tricyclic sesquiterpene derived from Pogostemon cablin of the Labiatae family, has demonstrated potential in modulating inflammatory responses and tumor progression. This study aimed to investigate the mechanisms through which patchouli alcohol influences inflammatory pathways associated with prostate cancer using bioinformatics and experimental validation.

Aims: Patchouli essential oil (PEO) is the major active ingredient of a famous medicinal plant Pogostemon cablin (Blanco) Benth. This study aims to investigate the anti-aging activities of PEO and its major component, and elucidate the underlying molecular mechanisms.

Main Methods: The anti-aging activities of PEO and its main component patchouli alcohol (PA) were investigated by examining the lifespan, senescence associated indicators as well as stress resistance of Caenorhabditis elegans.

Ethnopharmacological Relevance: Ulcerative colitis (UC), a chronic inflammatory bowel disease, has become a significant public health challenge due to the limited effectiveness of available therapies. Huoxiang Zhengqi (HXZQ), a well-established traditional Chinese formula, shows potential in managing UC, as suggested by clinical and pharmacological studies. However, the active components and mechanisms responsible for its effects remain unclear.

Article Synopsis
  • Patchouli alcohol (PA) is a special ingredient used in traditional Chinese medicine that can help with gut health.
  • A study found that PA can reduce inflammation in mice with ulcerative colitis (UC) by lowering levels of certain harmful substances in the body.
  • PA changes the makeup of gut bacteria but doesn't affect its overall diversity, and it helps protect the gut by targeting specific pathways related to inflammation.
