Article Abstract
A catalytic approach of synthesizing the -selective saturated carbo- and heterocyclic germanium compounds (3D framework) is reported the hydrogenation of readily accessible aromatic germanes (2D framework). Among the numerous catalysts tested, Nishimura's catalyst (RhO/PtO·HO) exhibited the best hydrogenation reactivity with an isolated yield of up to 96%. A broad range of substrates including the synthesis of unprecedented saturated heterocyclic germanes was explored. This selective hydrogenation strategy could tolerate several functional groups such as -CF, -OR, -F, -Bpin, and -SiR groups. The synthesized products demonstrated the applications in coupling reactions including the newly developed strategy of aza-Giese-type addition reaction (C-N bond formation) from the saturated cyclic germane product. These versatile motifs can have a substantial value in organic synthesis and medicinal chemistry as they show orthogonal reactivity in coupling reactions while competing with other coupling partners such as boranes or silanes, acquiring a three-dimensional structure with high stability and robustness.

Spirocyclic alkyl amino carbene (SCAAC) Ru complexes demonstrate outstanding activity and selectivity in ethenolysis of methyl oleate (MO) or fatty acid methyl esters (FAMEs), and 5,6-dimethoxyindane derivative was the most active catalyst to date. For the further catalyst design, we proposed modifying the spirocyclic fragment by fusion of saturated carbo- or heterocycle, linked to the 5,6-positions of indane or 6,7- positions of tetralin. Another suggested way of the modification of SCAAC complex was the insertion of chromane fragment to the carbene ligand.

The colony-stimulating factor 1 receptor (CSF1R) is an attractive target for inflammation disorders and cancers. Based on a series of pyrrolo[2,3-d]pyrimidine containing two carbo-aromatic rings, we have searched for new CSF1R inhibitors having a higher fraction of sp-atoms. The title compounds, CHNOS (), CHNOS (), and CHNOS (), form a homologous series in which the size of the saturated ring increases from six- to eight-membered (with four, five and six methyl-ene groups respectively). For and , the central (CH) moieties are all displaced to the same side of their ring, and the CH-CH-CH angles are much wider than the standard value; a database search indicates that these are general features of such ring systems. For , the thio-phene ring lies with the sulfur atom on the opposite side of the C-C bond to the cyano group, in contrast to and .
