The amide-I vibrational characteristics and conformational preferences of the model compound - histidine dipeptide (Ac-His-NHCH, HISD) in gas phase and solution have been revealed with the help of ab initio calculations and wavefunction analyses. The Gibbs free energy surfaces (FESs) of solvated HISD were smoothed by solvent effect to exhibit different structural populations concerning various external environments. It was shown that the most stable conformations of HISD in CHCl and gas phase are C7eq, while those in DMSO and water are β and PPII, respectively. Compared with ALAD, the number of accessible conformational states on these FESs was predicted to be reduced due to the steric effect of imidazole group. The two amide-I normal modes of HISD were found to have intrinsically secondary structural dependencies, and be sensitive to surrounding environments. The average amide-I frequencies of HISD isomers in these environments were predicted to be almost the same as those of ALAD, while the amide-I mean frequencies were estimated to be lower than ALAD due to the intramolecular interactions between the imidazole group and amino-terminal amide unit. The good linear correlations between amide-I frequencies and the atomic electrostatic potentials (ESPs) of amide groups were also found to interpret the solvent-induced amide-I frequency shifts of HISD at the electronic structure level. These results allow us to gain a deep understanding of amide-I vibrations of HISD, and would be helpful for the site-specific conformational monitoring and spectral interpretation of solvated polypeptides.

Quantum mechanical vibrational coherence transfer processes play important roles in energy relaxation, charge transfer, and reaction dynamics in chemical and biological systems but are difficult to directly measure using traditional condensed-phase nonlinear spectroscopies. Recently, we developed a new experimental capability to obtain two-dimensional infrared (2D IR) spectra of molecular systems in the gas phase that enables the direct measurement of coherence pathways. Herein, we report ultrafast 2D IR spectroscopy of the peptide glutathione (GSH) isolated and cryogenically cooled in the gas phase.

Sum frequency generation (SFG) necessitates both noncentrosymmetry and coherence over multiple length scales. These requirements make vibrational SFG spectroscopy capable of probing structural information of noncentrosymmetric organic crystals interspersed in polymeric matrices and their three-dimensional spatial distributions within the matrices without spectral interferences from the amorphous components. However, this analysis is not as straightforward as simple vibrational spectroscopy or scattering experiments; it requires knowing the molecular hyperpolarizability of SFG-active vibrational modes and their interplay within the coherence length.

Silk fibers have good biocompatibility and mechanical properties, which make them attractive in biomaterial applications as well as textile industries. It is believed that the superior mechanical property is associated with the crystalline β-sheet structure in the fiber; but a deeper understanding of the structure-property relationship is still needed for full exploitation of its physical properties. Especially, accurate information on hydrogen-bonding interactions within β-sheet domains at the nanoscale and their spatial distributions at the mesoscale are critically needed.
