Late-onset hypogonadism (LOH) refers to sexual and non-sexual symptoms in men caused by age-related decreases in circulating testosterone. Leydig cells (LCs) transplantation is considered to be one of a viable approach for LOH therapy, but the limited source of LCs limits the application of this approach. The aim of this study was to induce the directed differentiation of rat bone marrow mesenchymal stem cells (BMSCs) into LCs in vitro, and explore the potential involvement of Wnt/β-catenin signaling pathway in the differentiation process.

Objectives: We evaluated potential therapeutic efficacy of LGK-974 and ETC-159 in fibrotic scleroderma cells.

Methods: Primary scleroderma dermal fibroblast cells of mouse origin (SSc fibroblasts) and primary fibrotic lung fibroblast cells of human origin (CCL-191) were used in this study. PORCN inhibitors LGK-974 (S7143, 1 μM; Selleckchem, USA) and ETC-159 (S7143, 10 μM; Selleckchem, USA) were used.

Department of Gastrointestinal Surgery, Hainan General Hospital, Hainan Affiliated Hospital of Hainan Medical University, Hainan, Haikou, Hainan Province, People's Republic of China.

Article Synopsis
  • * Overexpressing WNT7B increases glucose uptake and lactic acid production in colon cancer cells by upregulating GLUT1, while knocking it down reverses these effects.
  • * Inhibition of the WNT/β-catenin pathway leads to decreased GLUT1 levels and increased apoptosis, indicating that WNT7B promotes tumor growth through enhanced glucose metabolism and resistance to cell death.

Elevated Porcupine Disrupts Lipid Metabolism and Promotes Inflammatory Response in MASLD.

Liver Int

October 2024

Department of Pharmacology and School of Basic Medicine Clinical Pharmacy, China Pharmaceutical University, Nanjing, Jiangsu, China.

Article Synopsis
  • Metabolic dysfunction-associated steatotic liver disease (MASLD) is a common condition linked to serious liver issues, with high serum levels of palmitic acid (PA) but unclear connections between PA and MASLD development.
  • The study utilized advanced methods like gene expression analysis and animal models to investigate the role of the protein Porcupine in MASLD, finding that PA induces Porcupine production leading to inflammation and altered lipid metabolism.
  • Results indicate that targeting Porcupine with an inhibitor called Wnt974 can reduce lipid buildup and inflammation in the liver, suggesting Porcupine as a potential therapeutic target for MASLD.

Increased oxygen stimulation promotes chemoresistance and phenotype shifting through PLCB1 in gliomas.

September 2024

Glioma Medical Research Center and Department of Neurosurgery, The First Affiliated Hospital, Third Military Medical University (Army Medical University), Chongqing 400038, China. Electronic address:

Gliomas, the most common CNS (central nerve system) tumors, face poor survival due to severe chemoresistance exacerbated by hypoxia. However, studies on whether altered hypoxic conditions benefit for chemo-sensitivity and how gliomas react to increased oxygen stimulation are limited. In this study, we demonstrated that increased oxygen stimulation promotes glioma growth and chemoresistance.
