Background: Platelet glycoprotein (GP) Ibα is the major ligand-binding subunit of the GPIb-IX-V complex that binds von Willebrand factor. GPIbα is heavily glycosylated, and its glycans have been proposed to play key roles in platelet clearance, von Willebrand factor binding, and as target antigens in immune thrombocytopenia syndromes. Despite its importance in platelet biology, the glycosylation profile of GPIbα is not well characterized.
Objectives: The aim of this study was to comprehensively analyze GPIbα amino acid sites of glycosylation (glycosites) and glycan structures.
Methods: GPIbα ectodomain that was recombinantly expressed or that was purified from human platelets was analyzed by Western blot, mass spectrometry glycomics, and mass spectrometry glycopeptide analysis to define glycosites and the structures of the attached glycans.
Results: We identified a diverse repertoire of N- and O-glycans, including sialoglycans, Tn antigen, T antigen, and ABO(H) blood group antigens. In the analysis of the recombinant protein, we identified 62 unique O-glycosites. In the analysis of the endogenous protein purified from platelets, we identified 48 unique O-glycosites and 1 N-glycosite. The GPIbα mucin domain is densely O-glycosylated. Glycosites are also located within the macroglycopeptide domain and mechanosensory domain.
Conclusions: This comprehensive analysis of GPIbα glycosylation lays the foundation for further studies to determine the functional and structural roles of GPIbα glycans.

This study shows the efficiency of WH-C450, an adsorbent obtained from water hyacinth (WH) biomass, in the removal of sulfamethoxazole (SMX) from aqueous solutions. The process involves calcination of WH at 450 °C to produce an optimal adsorbent material capable of removing up to 73% of SMX and maximum SMX adsorption capacity of 132.23 mg/g.

Vision transformer (ViT)and convolutional neural networks (CNNs) each possess distinct strengths in medical imaging: ViT excels in capturing long-range dependencies through self-attention, while CNNs are adept at extracting local features via spatial convolution filters. While ViT may struggle with capturing detailed local spatial information, critical for tasks like anomaly detection in medical imaging, shallow CNNs often fail to effectively abstract global context. This study aims to explore and evaluate hybrid architectures that integrate ViT and CNN to leverage their complementary strengths for enhanced performance in medical vision tasks, such as segmentation, classification, reconstruction, and prediction.

Analysis of the symmetry of the brain hemispheres at the level of individual structures and dominant tissue features has been the subject of research for many years in the context of improving the effectiveness of imaging methods for the diagnosis of brain tumor, stroke, and Alzheimer's disease, among others. One useful approach is to reliably determine the midline of the brain, which allows comparative analysis of the hemispheres and uncovers information on symmetry/asymmetry in the relevant planes of, for example, CT scans. Therefore, an effective method that is robust to various geometric deformations, artifacts, varying noise characteristics, and natural anatomical variability is sought.

The ecological niche separation of microbial interactions in forest ecosystems is critical to maintaining ecological balance and biodiversity and has yet to be comprehensively explored in microbial ecology. This study investigated the impacts of soil properties on microbial interactions and carbon metabolism potential in forest soils across 67 sites in China. Using redundancy analysis and random forest models, we identified soil pH and dissolved organic matter (DOM) aromaticity as the primary drivers of microbial interactions, representing abiotic conditions and resource niches, respectively.
