In addition to its canonical function in protecting from pathogens, the immune system can also promote behavioural alterations . The scope and mechanisms of behavioural modifications by the immune system are not yet well understood. Using a mouse food allergy model, here we show that allergic sensitization drives antigen-specific behavioural aversion. Allergen ingestion activates brain areas involved in the response to aversive stimuli, including the nucleus of tractus solitarius, parabrachial nucleus, and central amygdala. Food aversion requires IgE antibodies and mast cells but precedes the development of gut allergic inflammation. The ability of allergen-specific IgE and mast cells to promote aversion requires leukotrienes and growth and differentiation factor 15 (GDF15). In addition to allergen-induced aversion, we find that lipopolysaccharide-induced inflammation also resulted in IgE-dependent aversive behaviour. These findings thus point to antigen-specific behavioural modifications that likely evolved to promote niche selection to avoid unfavourable environments.

The number of opioid overdose deaths has increased over the past several years, mainly driven by an increase in the availability of highly potent synthetic opioids, like fentanyl, in the un-regulated drug supply. Over the last few years, changes in the drug supply, and in particular the availability of counterfeit pills containing fentanyl, have made oral use of opioids a more common route of administration. Here, we used a drinking in the dark (DiD) paradigm to model oral fentanyl self-administration using increasing fentanyl concentrations in male and female mice over 5 weeks.

Background: Post-traumatic stress disorder (PTSD) causes intrusive symptoms and avoidance behaviours due to dysregulation in various brain regions, including the hippocampus. Deep brain stimulation (DBS) shows promise for refractory PTSD cases. In rodents, DBS improves fear extinction and reduces anxiety-like behaviours, but its effects on active-avoidance extinction remain unexplored.

In this article, we use the example of pain exposure therapy to illustrate how behavioral pain treatments can be systematically personalized following the principles of functional analysis. Based on the fear-avoidance model, pain exposure therapy has evolved as a mechanistically-based treatment to modify the mechanism of avoidance learning with the aim to reduce disability levels. We first present experimental evidence on avoidance learning from a general psychological perspective.
