The objective of the study was to develop a new method for the determination of the total content of vitamin C and dehydroascorbic acid in food, based on the technique of differential pulse voltammetry with the use of a boron-doped diamond electrode modified with mercury film. A comparison was made between the results obtained with the developed method and a proposed reference method based on high-performance liquid chromatography with spectrophotometric detection. The reduction of dehydroascorbic acid was performed with the use of tris(2-carboxyethyl)phosphine. The interference caused by the presence of tris(2-carboxyethyl)phosphine during the voltammetric determination of ascorbic acid was effectively eliminated through a reaction with N-ethylmaleimide. The conducted validation of the voltammetric method indicated that correct results of analysis of the total content of vitamin C and ascorbic acid were obtained. Analysis of the content of dehydroascorbic acid was imprecise due to the application of the differential method. The results of the analyses and the determined validation parameters of the developed method are characterised by a high degree of conformance with the results obtained with the chromatographic reference method, which indicates the equivalence of the two methods.

Anticancer effects of high-dose vitamin C (VC) have been evaluated on many cancer cell lines, and its efficacy in clinical trials and in combination with anticancer drugs or radiation have been reported; however, its effect on gastric cancer and its mechanisms remain unclear. In the present study, the cell growth inhibitory/lethal effects of high-dose ascorbic acid (AsA), a reduced form of VC was examined on three gastric cancer cell lines. Of these, signet ring cell carcinoma NUGC-4 cells were the most sensitive, but the effects were small and limited in normal cells.

Cadmium stress (CS) induced the peroxide damage and inhibited wheat photosynthetic capacity and growth. Compared to CS, selenium (Se) application plus CS bolstered chlorophyll and carotenoid contents, photosynthetic rate, the maximum photochemical efficiency of PSII, the quantum yield of PSII photochemistry, and photochemical quenching, superoxide dismutase, catalase, ascorbate peroxidase, glutathione reductase, L-galactono-1,4-lactone dehydrogenase, and gamma-glutamylcysteine synthetase activities, ascorbic acid and glutathione contents, AsA/dehydroascorbic acid and GSH/oxidized glutathione, and decreased nonphotochemical quenching (q), antioxidant biomarkers malondialdehyde and hydrogen peroxide contents, and electrolyte leakage (EL). At the same time, Se alone declined antioxidant biomarkers contents, q and EL, and augmented the rest of the aforementioned indexes.

Article Synopsis
  • Current chemical drugs are ineffective for tick control and there are no effective vaccines, leading to the urgent need for new control strategies.
  • Small interfering RNAs (siRNAs) have potential to help control parasites, but their effectiveness is limited by challenges in getting them to the RNA-induced silencing complex (RISC).
  • This study developed a delivery system using chitosan nanoparticles to successfully transfect siRNAs into parasites, which reduced target gene expression and inhibited parasite growth, opening new avenues for gene therapy and understanding host-parasite interactions.

Dehydroascorbic acid quantification in human plasma: Simultaneous direct measurement of the ascorbic acid/dehydroascorbic acid couple by UPLC/MS-MS.

Ascorbic acid (AA, vitamin C) and dehydroascorbic acid (DHA) constitute a biological couple. No technique can accurately, independently, and simultaneously quantify both members of the couple in animal and human samples, thereby constraining advances in physiology and pathophysiology. Here we describe a new UPLC/MS/MS method to measure both compounds directly and independently in human plasma.
