Hyaluronan is a non-sulfated glycosaminoglycan synthesized on the plasma membrane of almost all mammalian cells, which can interact with different proteoglycans of the extracellular matrix. Aggrecan, versican, neurocan, and brevican are proteoglycans whose structures present a specific protein domain called "link module," which allows hyaluronan binding. Therefore, they can be helpful for assays that detect hyaluronan. For example, ELISA-like methods developed to measure hyaluronan amounts in solution are based on specific interactions between this molecule and the link module present in aggrecan or other hyaluronan-binding proteins (hyaladherins).

Chronic immunoinflammatory rheumatic diseases, such as axial spondyloarthritis (AxSpA), are accompanied by a dysregulation of bone remodeling. Among potential biomarkers of bone metabolism, the Wnt pathway antagonist, Dickkopf-1 (DKK-1), is of particular interest because of its potential to reflect a shift towards joint ossification or osteoporosis, but its diagnostic value needs validation. There is still a lack of stable and efficient methods of measuring serum DKK-1 levels suitable for longitude studies.

Article Synopsis
  • Hyaluronan (HA) is an important molecule involved in various biological processes and has implications in health and disease, such as tissue hydration and tumor growth.
  • Traditional methods for measuring HA often lack sensitivity and specificity, particularly for lower molecular weights, which limits their effectiveness.
  • This study presents a new method using LC-MS/MS, capable of detecting HA at very low levels and successfully applied it to analyze HA levels in mouse tissues, showing significant changes in response to liver injury.

Characteristics of Hyaluronan Metabolism During Myofibroblast Differentiation in Orbital Fibroblasts.

Invest Ophthalmol Vis Sci

November 2024

Division of Endocrinology, Department of Internal Medicine, Faculty of Medicine, University of Debrecen, Debrecen, Hungary.

Article Synopsis
  • The study investigates how myofibroblast differentiation (MD) affects hyaluronan (HA) turnover in orbital fibroblasts, particularly in the context of thyroid eye disease (TED).
  • Using primary cultures from TED and non-TED tissue samples, researchers induced MD and measured various factors like proliferation, HA levels, and enzyme expressions related to HA metabolism.
  • Findings revealed that MD leads to lower HA turnover by inhibiting certain enzymes while increasing HA synthesis, suggesting that targeting specific hyaluronidases could be a new approach to treat TED.

A sensitive assay for measuring whole-blood responses to type I IFNs.

Proc Natl Acad Sci U S A

October 2024

Laboratory of Human Genetics of Infectious Diseases, Necker Branch, Institut National de la Santé et de la Recherche Médicale U1163, Necker Hospital for Sick Children, Paris 75015, France.

Article Synopsis
  • Inborn errors or autoantibodies (auto-Abs) against type I interferons (IFNs) can lead to severe viral infections.
  • Researchers developed a straightforward blood test that can identify these conditions by stimulating blood with glycosylated IFN-α2, -β, or -ω and measuring IP-10 levels.
  • The study found that IP-10 levels in patients with inherited deficiencies only increase with type II IFN (IFN-γ), while those with auto-Abs can still respond to non-neutralized type I IFNs.

Amateurs often struggle with detecting and quantifying protein biomarkers in body fluids due to the high expertise required. This study introduces a Lab-in-a-Vial (LV) rapid diagnostic platform, featuring hydrangea-like platinum nanozymes (PtNH), for rapid, accurate detection and quantification of protein biomarkers on-site within 15 min. This method significantly enhances detection sensitivity for various biomarkers in body fluids, surpassing traditional methods such as enzyme-linked immunosorbent assays (ELISA) and lateral flow assays (LFA) by ≈250 to 1300 times.
