In health, the gut microbiome functions as a stable ecosystem maintaining overall balance and ensuring its own survival against environmental stressors through complex microbial interaction. This balance and protection from stressors is maintained through interactions both within the bacterial ecosystem as well as with its host. As a consequence, the gut microbiome plays a critical role in various physiological processes including maintaining the structure and function of the gut barrier, educating the gut immune system, and modulating the gut motor, digestive/absorptive, as well as neuroendocrine system all of which are crucial for human health and disease pathogenesis.

Host-microbiota cooperation is critical for successful intestinal homeostasis. The commensal segmented filamentous bacteria (SFB) are crucial for orchestrating the post-natal maturation of the host gut immune system and establishing a healthy state of physiological inflammation, which largely depends on their intimate attachment to the ileal mucosa. However, the signaling pathways used by SFB to induce gut immune responses and how such responses ultimately control SFB colonization remain controversial.

This study aims to investigate the pathogenesis of depression in mice using the chronic unpredictable mild stress (CUMS) model, with a particular focus on the changes in inflammatory gene networks and inflammatory factor levels under the condition of gut microbiota dysbiosis. The results indicate that CUMS-induced mice exhibited significant depressive-like behaviors. Specifically, they displayed reduced sucrose intake in the sucrose preference test, decreased central area distance and time in the open field test, and reduced percentage of entries and time spent in the open arm in the elevated plus maze test.
