Emerging evidence shows that the biomechanical environment is required to support cancer stem cells (CSCs), which play a crucial role in drug resistance. However, how mechanotransduction signals regulate CSCs and its clinical significance has remained unclear. Using clinical-practice ultrasound elastography for patients' lesions and atomic force microscopy for surgical samples, we reveal that increased matrix stiffness is associated with poor responses to neoadjuvant chemotherapy, worse prognosis, and CSC enrichment in patients with breast cancer. Mechanically, TAZ activated by biomechanics enhances CSC properties via phase separation with NANOG. TAZ-NANOG phase separation, which is dependent on acidic residues in the N-terminal activation domain of NANOG, promotes the transcription of SOX2 and OCT4. Therapeutically, targeting NANOG or TAZ reduces CSCs and enhances the chemosensitivity in vivo. Collectively, this study demonstrated that the phase separation of a pluripotency transcription factor links mechanical cues in the niche to the fate of CSCs.

Biphasic system not only presents a promising opportunity for complex catalytic processes, but also is a grand challenge in efficient tandem reactions. As an emerging solar-to-chemical conversion, the visible-light-driven and water-donating hydrogenation combines the sustainability of photocatalysis and economic-value of hydrogenation. However, the key and challenging point is to couple water-soluble photocatalytic hydrogen evolution reaction (HER) with oil-soluble hydrogenation.

The protein corona effect refers to the phenomenon wherein nanomaterials in the bloodstream are coated by serum proteins, yet how protein coronated nanomaterials interact with blood vessels and its toxicity implications remain poorly understood. In this study, we investigated protein corona-related vessel toxicity by using an all-humanized assay integrating blood vessel organoids and patient-derived serum. Initially, we screened various nanomaterials to discern how parameters including size, morphology, hydrophobicity, surface charge, and chirality-dependent protein corona difference influence their uptake by vessel organoids.

Nucleocapsid assembly drives Ebola viral factory maturation and dispersion.

Schaller Research Groups, Department of Infectious Diseases, Virology, Heidelberg University, Heidelberg, Germany; BioQuant, Heidelberg University, Heidelberg, Germany. Electronic address:

Replication and genome encapsidation of many negative-sense RNA viruses take place in virus-induced membraneless organelles termed viral factories (VFs). Although liquid properties of VFs are believed to control the transition from genome replication to nucleocapsid (NC) assembly, VF maturation and interactions with the cellular environment remain elusive. Here, we apply in situ cryo-correlative light and electron tomography to follow NC assembly and changes in VF morphology and their liquid properties during Ebola virus infection.

Formaldehyde-purifying air conditioner with integrated gas-phase photoelectrocatalytic system.

Developing an efficient and economical indoor air purification system for catalytic decomposition of formaldehyde is of great significance. In this work, an indoor air conditioner capable of purifying formaldehyde was designed by directly integrating defective WO/TiO nanotube catalytic fin, with both thermal conductivity and gas-phase photoelectrocatalytic (GPEC) properties, onto the condenser component. The electrochemical treatment of the catalytic fin introduced a substantial number of oxygen vacancies, resulting in a significant increase in carrier concentration and mobility to the semi-metallic level.
