Malignant tumor tissues exhibit inter- and intratumoral heterogeneities, aberrant development, dynamic stromal composition, diverse tissue phenotypes, and cell populations growing within localized mechanical stresses in hypoxic conditions. Experimental tumor models employing engineered systems that isolate and study these complex variables using techniques are under development as complementary methods to preclinical models. Here, advances in extrusion bioprinting as an enabling technology to recreate the three-dimensional tumor milieu and its complex heterogeneous characteristics are reviewed. Extrusion bioprinting allows for the deposition of multiple materials, or selected cell types and concentrations, into models based upon physiological features of the tumor. This affords the creation of complex samples with representative extracellular or stromal compositions that replicate the biology of patient tissue. Biomaterial engineering of printable materials that replicate specific features of the tumor microenvironment offer experimental reproducibility, throughput, and physiological relevance compared to animal models. In this review, we describe the potential of extrusion-based bioprinting to recreate the tumor microenvironment within models.

Bioprinting and bioinks are two of the game changers in bone tissue engineering. This review presents different bioprinting technologies including extrusion-based, inkjet-based, laser-assisted, light-based, and hybrid technologies with their own strengths and weaknesses. This review will aid researchers in the selection and assessment of the bioink; the discussion ranges from commercially available bioinks to custom lab-made formulations mainly based on natural polymers, such as agarose, alginate, gelatin, collagen, and chitosan, designed for bone tissue engineering.

Bioprinting a resilient yet optically transparent corneal tissue substitute remains a challenge. In this study we introduce an innovative methodology aimed at bolstering the mechanical and optical attributes of silk fibroin (SF) hydrogels, pivotal for the progression of cornea tissue engineering. We devised a unique eosin Y-based photoinitiator system to instigate di-tyrosine linkages within highly concentrated pristine SF solutions under green light exposure.

3D bioprinting can generate the organized structures found in human skin for a variety of biological, medical, and pharmaceutical applications. Challenges in bioprinting skin include printing different types of cells in the same construct while maintaining their viability, which depends on the type of bioprinter and bioinks used. This study evaluated a novel 3D bioprinted skin model containing human keratinocytes (HEKa) and human dermal fibroblasts (HDF) in co-culture (CC) using a high-viscosity fibrin-based bioink produced using the BioX extrusion-based bioprinter.
