Characterization of jacalin, the human IgA and IgD binding lectin from jackfruit.

Published: May 1987

The lectin jacalin from the jackfruit Artocarpus heterophyllus reacts by precipitation and western blotting with human IgA1 and IgD but not with IgA2 (nor IgG and IgM). However, it weakly binds IgA2 as shown by affinity chromatography and competitive ELISA. Predominantly reactive carbohydrates are D-galactose and N-acetyl D-galactosamine. Jacalin has an apparent Mr of about 54,000 and is probably made up of three non-glycosylated and one glycosylated non-covalently linked subunits. Its electrophoretic properties and amino acid and carbohydrate composition are indicated.

Comparative analysis of the gene family in the whole-genome of five gramineous plants.

Front Plant Sci

December 2024

Department of Plant Sciences, College of Life Sciences, Wuhan University, Wuhan, China.

The Jacalin-related lectins () gene family play a crucial role in regulating plant development and responding to environmental stress. However, a systematic bioinformatics analysis of the gene family in Gramineae plants has been lacking. In this study, we identified 101 JRL proteins from five Gramineae species and classified them into eight distinct clades.

Evaluation of Jacalin lectin sorbents for the extraction of the human chorionic gonadotropin glycoforms prior to analysis by nano liquid chromatography-high resolution mass spectrometry.

Human chorionic gonadotropin (hCG) is a dimeric, highly glycosylated hormone with a total of 4 N- and 4 O-glycosylation sites in its two subunits, hCGα and hCGβ. Recently, we developed a novel nano liquid chromatography coupled to high resolution mass spectrometry (nanoLC-HRMS) method for the analysis and thus the detection of the intact glycoforms of hCG. Here, a sorbent functionalized with the Jacalin lectin was evaluated in solid-phase extraction (SPE) for its potential to fractionate the hCG glycoforms prior to their nanoLC-HRMS analysis at the intact level, which may facilitate the detection of low-abundance glycoforms and may lead to a more detailed characterization of the hormone glycosylation.

The jacalin-related lectins (JRLs) are widely distributed in plants and are involved in plant development and multiple stress responses. However, the characteristics of the gene family at the genome-wide level and the roles of JRLs in barley's response to low-nitrogen (LN) stress have been rarely reported. In this study, 32 genes were identified and unevenly distributed at both ends of the seven chromosomes in barley.

Assembly and recognition mechanisms of glycosylated PEGylated polyallylamine phosphate nanoparticles: A fluorescence correlation spectroscopy and small angle X-ray scattering study.

J Colloid Interface Sci

September 2023

Soft Matter Nanotechnology Group, CIC biomaGUNE, Basque Research and Technology Alliance (BRTA), Paseo Miramón 182, 20014 Donostia-San Sebastián, Guipúzcoa, Spain. Electronic address:

Hypothesis: Modification of polyallylamine hydrochloride (PAH) with heterobifunctional low molecular weight polyethylene glycol (PEG) (600 and 1395 Da), and subsequent attachment of mannose, glucose, or lactose sugars to PEG, can lead to formation of polyamine phosphate nanoparticles (PANs) with lectin binding affinity and narrow size distribution.

Experiments: Size, polydispersity, and internal structure of glycosylated PEGylated PANs were characterized by transmission electron microscopy (TEM), dynamic light scattering (DLS) and small angle X-ray scattering (SAXS). Fluorescence correlation spectroscopy (FCS) was used to study the association of labelled glycol-PEGylated PANs.

Lectins are proteins that reversibly bind to carbohydrates and are commonly found across many species. The Banana Lectin (BanLec) is a member of the Jacalin-related Lectins, heavily studied for its immunomodulatory, antiproliferative, and antiviral activity. In this study, a novel sequence was generated in silico considering the native BanLec amino acid sequence and 9 other lectins belonging to JRL.
