In the chalcogen series, tellurium species exhibit the strongest chalcogen bonding (ChB) interaction with electron-rich atom. This property explains the renewed interested toward tellurium-based derivatives and their use in different applications, such as organocatalysis. In this context, the catalytic activity of telluronium salts in the Povarov reaction is presented herein. Different dienophiles, as well as imines of variable electronic nature, efficiently react in the presence of catalytic amount of either diarylmethyltelluronium or triaryltelluronium salts. Both catalysts could also readily perform the three-component Povarov reaction starting from aldehyde, aniline and dihydrofuran. The reactivity of telluroniums towards imines and aldehydes was confirmed in the solid state by the ability of Te atom to interact through ChB with the oxygen carbonyl of acetone, and in solution with significant shift variations of the imine proton and of the tellurium atom in H and Te NMR spectroscopy. For the most active telluronium catalysts bearing CF groups, association constants (K) with N-phenyl phenylmethanimine in the range 22-38 M were measured in dichloromethane.

A straightforward I2-promoted Povarov multicomponent reaction strategy for the synthesis of N-heteroaromatic benzothiazole and 2, 2'-biquinolines was described. This methodology involved a one-pot two-step procedure, tandem iodination/Kornblum oxidation, and [4 + 2] condensation. This protocol was characterized by broad scope of substrates (49 examples, 31-80% yields), applicable for large-scale preparation, and downstream derivatization of the product.

The protein kinases CLK and ROCK play key roles in cell growth and migration, respectively, and are potential anticancer targets. ROCK inhibitors have been approved by the FDA for various diseases and CLK inhibitors are currently being trialed in the clinic as anticancer agents. Compounds with polypharmacology are desired, especially in oncology, due to the potential for high efficacy as well as addressing resistance issues.

The synthesis of phosphorous indenoquinolines and their biological evaluation as topoisomerase 1 (TOP1) inhibitors and antiproliferative agents were performed. First, the preparation of new hybrid 5-indeno[2,1-]quinolines with a phosphine oxide group was performed by a two-step Povarov-type [4+2]-cycloaddition reaction between the corresponding phosphorated aldimines with indene in the presence of BF·EtO. Subsequent oxidation of the methylene present in the structure resulted in the corresponding indeno[2,1-]quinolin-7-one phosphine oxides .

In this contribution, a novel, simple, diastereoselective and environmentally benign two-step diversity-oriented synthesis of imidazo[4,5,1-]quinolines is described for the first time. The synthesis of the target compounds involves a deep eutectic solvent-mediated one-pot Povarov reaction leading to the obtention of 8-nitrotetrahydroquinolines, followed by a microwave-assisted reductive cyclocondensation employing different aromatic and aliphatic aldehydes. The target compounds were obtained in up to 70% overall yield starting from commercially available -nitroanilines, natural phenylpropanoids (-anethole and -isoeugenol) and aromatic or aliphatic aldehydes.

Aza-Diels-Alder cycloaddition reaction is a critical synthetic method for the production of bioactive tetrahydroquinolines. To this aim, an imine obtained from the reaction of an aniline derivative and a carbonyl compound is cyclized with an alkene in the presence of a catalyst. In this research, some tetrahydroquinoline compounds are synthesized through aza-Diels-Alder reaction in the presence of a prepared Ce(III) immobilized on the functionalized halloysite (Ce/Hal-TCT-IDA) as a catalyst.
