A semisynthetic electrocatalyst for carbon dioxide reduction to carbon monoxide in water is reported. Cobalt microperoxidase-11 (CoMP11-Ac) is shown to reduce CO to CO with a turnover number of up to 32,000 and a selectivity of up to 88:5 CO:H. Higher selectivity for CO production is favored by a less cathodic applied potential and use of a higher p buffer. A mechanistic hypothesis is presented in which avoiding the formation and protonation of a formal Co(I) species favors CO production. These results demonstrate how tuning reaction conditions impact reactivity toward CO reduction for a biocatalyst previously developed for H production.

Oxygen scavenging systems (OSSs) are critical for dye stability in single-molecule fluorescence (SMF) experiments. However, the commonly used protocatechuic acid (PCA)/protocatechuate-3,4-dioxygenase (PCD) OSS alters DNA mechanical properties, limiting its applicability. To address this limitation, we examine the bilirubin oxidase (BOD) OSS, which had not been previously used in single-molecule experiments, alongside the pyranose oxidase and catalase (POC) OSS.

The production of hydrogen by water electrolysis benefits from the development of water oxidation catalysts. This development process can be aided by the postulation of design rules for catalytic systems. The analysis of the reactivity of molecular complexes can be complicated by their decomposition under catalytic conditions into nanoparticles that may also be active.

Multi-attribute PAT for UF/DF of Proteins-Monitoring Concentration, particle sizes, and Buffer Exchange.

April 2020

Institute of Engineering in Life Sciences, Section IV: Biomolecular Separation Engineering, Karlsruhe Institute of Technology, Fritz-Haber-Weg 2, 76131, Karlsruhe, Germany.

Ultrafiltration/diafiltration (UF/DF) plays an important role in the manufacturing of biopharmaceuticals. Monitoring critical process parameters and quality attributes by process analytical technology (PAT) during those steps can facilitate process development and assure consistent quality in production processes. In this study, a lab-scale cross-flow filtration (CFF) device was equipped with a variable pathlength (VP) ultraviolet and visible (UV/Vis) spectrometer, a light scattering photometer, and a liquid density sensor (microLDS).

Fast Ca Buffer-Dependent Reliable but Plastic Transmission at Small CNS Synapses Revealed by Direct Bouton Recording.

The small size of presynaptic structures and their rapid function have obscured the mechanisms underlying neurotransmission and plasticity. To dissect the function of conventional small presynaptic boutons, we performed direct recording using axon varicosities of cerebellar granule cells (GCs), a parallel-fiber bouton, in dissociated culture, in which pre- and postsynaptic paired recordings are feasible. Identification and accessibility of EGFP-labeled GC boutons allowed us to patch-clamp record presynaptic voltage-gated Ca currents and membrane capacitances, together with excitatory postsynaptic currents.
