The stereodivergent synthesis of - and -2,6-disubstituted tetrahydropyrans (THPs) via sodium hexamethyldisilazide-promoted oxa-Michael cyclization of ()-ζ-hydroxy α,β-unsaturated esters is presented. The cyclization affords the kinetically favored -THPs with high stereoselectivity (dr up to 93:7) at a low temperature (-78 °C), while the room-temperature reaction does not produce the thermodynamically preferred -THPs as major products and occurs with poor stereocontrol. The addition of tetramethylethylenediamine (TMEDA) significantly improves the stereochemical outcome of the room-temperature cyclization and allows attaining high -selectivity (dr up to 99:1). The remarkable effect of TMEDA indicates that the sodium cation plays an important role in controlling the stereoselectivity of the thermodynamically driven process, that is, complexation of the cation with the cyclization products results in diminished selectivity. DFT calculations support this conclusion, indicating a greater difference in Gibbs energies of sodium-free - and -enolates compared to the respective sodium chelate complexes. The synthetic utility of the method has been demonstrated by the formal syntheses of (+)-Neopeltolide and (-)-Diospongin B and the total synthesis of (-)-Diospongin A.

Herein, we disclose a cinchona-alkaloid-based squaramide-catalyzed intermolecular oxa-Michael cascade addition cyclization of -protected hydroxyl amine to formyl-tethered Michael acceptors toward the synthesis of enantioenriched 1,2-oxazine scaffolds. Generally, good yield (up to 78%), good to excellent enantiomeric ratio (up to 98:2 er), and excellent regioselectivity (>19:1) were observed. Furthermore, the scalability of this methodology and a successful demonstration of postsynthetic transformations have been accomplished.

The umpolung of aldimines using N-heterocyclic carbenes (NHCs) is less explored compared to the established polarity reversal of aldehydes. Described herein is an NHC-catalyzed imine umpolung /6π-electrocyclization cascade, which leads to the atom- and pot-economic synthesis of biologically important dihydrochromeno indoles. For the first time, the nucleophilic aza-Breslow intermediates have been intercepted with unactivated alkynes.

It was recognized only recently that the sister norcembranoids scabrolides A and B have notably different carbotricyclic scaffolds. Therefore, our synthesis route leading to scabrolide A could not be extended to its sibling. Reported is the first asymmetric total synthesis of sarglamides A, C, D, E, and F within 7 steps, featuring an intermolecular Diels-Alder cycloaddition of ()-phellandrene and 1,4-benzoquinone and intramolecular (-)Michael addition to construct the tetracyclic core of sarglamides. Importantly, our work demonstrated that the hypothetic Diels-Alder reaction of α-phellandrene with dienophile toussaintine C (or analogues) originally proposed as a biosynthetic pathway was not viable under non-enzymatic conditions.
