Five undescribed polyketide derivatives, pestaloketides A-E (-), along with eleven known analogues (-), were isolated from the sponge-derived fungus sp. Their structures, including absolute configurations, were elucidated by analyses of NMR spectroscopic HRESIMS data and electronic circular dichroism (ECD) calculations. Compounds , , , and exhibited weak cytotoxicities against four human cancer cell lines, with IC values ranging from 22.1 to 100 μM. Pestaloketide A () is an unusual polyketide, featuring a rare 5/10/5-fused ring system. Pestaloketides A () and B () exhibited moderately inhibited LPS-induced NO production activity, with IC values of 23.6 and 14.5 μM, respectively, without cytotoxicity observed. Preliminary bioactivity evaluations and molecular docking analysis indicated that pestaloketides A () and B () had the potential to be developed into anti-inflammatory activity drug leads.

Ketoreductases (KRs) are domains in the reductive loops of type I polyketide synthases (PKSs) and are responsible for the majority of stereocenters in reduced polyketides. Although the highly stereoselective reduction of ACP-bound β-ketothioester intermediates by KRs is crucial for the overall functioning of PKSs, the substrate-dependent stereoselectivity of KRs is a factor that is not yet fully understood, especially for KR domains in late PKS modules that act on biosynthetic precursors with complex polyketidic moieties. We present studies on the three KR domains FosKR7, PlmKR6, and EryKR6 from the biosynthetic pathways of fostriecin, phoslactomycin, and erythromycin by in vitro assays using close surrogates of the octaketidic FosKR7 biosynthetic precursor, complex derivatives and a diketide in the form of their biomimetic -acetylcysteamine thioesters.

Broad-spectrum crop protection technologies, such as abamectin and bifenthrin, are globally relied upon to curb the existential threats from economic crop pests such as the generalist herbivore Koch (TSSM).
