The design of hypoallergenic derivatives is a new strategy for allergen-specific immunotherapy. Although hypoallergenic derivatives of (mud crab) heat-stable tropomyosin (TM) and myosin light chain (MLC) have been preliminarily explored, their allergenicity needs to be further studied. In this work, recombinant allergens (wtTM, wtMLC) and hypoallergenic derivatives (mtTM, mtMLC) were purified. IgE-binding frequencies of wtTM and wtMLC in 177 crab-sensitised patients were 32.8% and 11.9%, respectively. In the Balb/c mouse model, mtTM and mtMLC caused mild intestinal inflammation, did not activate T-helper (Th) 2 immune response (interleukin-4, anaphylactic mediator, IgE, and IgG1 antibodies were not significantly increased) but could significantly promote the production of interleukin-10, which equilibrated Th1/Th2 cells, thus alleviating allergic symptoms. Moreover, mtTM and mtMLC-induced rabbit/mice anti-IgG antibodies could effectively block wtTM and wtMLC binding to patients' sera IgE . These results indicate that hypoallergenic derivatives offer the promise for an immunotherapeutic regimen for crab allergy.

Panthenol, a vitamin B5 derivative, is a key precursor in the synthesis of fatty acids that maintain epithelial function. Its ability to aid in moisturization, skin barrier restoration, and wound healing has led to its frequent incorporation into personal care products and pharmaceuticals-many of which are marketed to be hypoallergenic. While contact allergy to panthenol is rather uncommon, rates of positive reactions are on the rise, increasing from 0.

The ingestion of red kidney bean products is hindered by the persistent allergenicity of lectins, even after autoclaving. This study examined the modification of lectin allergenicity in red kidney beans by pH-shifting and autoclaving treatments, utilizing BALB/c mouse sensitization, recirculating perfusion, and a bone marrow-derived dendritic cell (BMDC) model for allergenicity evaluation. Compared to autoclaving alone, combined pH-shifting and autoclaving reduced allergic symptoms in BALB/c mice, as evidenced by lower serum IgE, mMCPT-1, GM-CSF, HIS, IL-2, IL-4, IL-9, IL-13, and IL-17 levels and higher IgG1, IgG2a, IL-10, IFN-γ, and IFN-α cytokine release.

Introduction: House dust mite is the primary trigger of allergic respiratory diseases worldwide, and allergen-specific immunotherapy (AIT) is the only disease-modifying treatment in the clinic. The use of allergen molecules instead of extracts is a promising strategy in AIT. In this study, we constructed a peptide hybrid vaccine against the major mite allergen Der f 35 and verified its hypoallergenicity, making it to be a promising candidate for AIT of mite allergy.

Pru p 3, a member of the lipid transfer protein family, is considered a major allergen from peach as it often induces serious allergic reactions in peach-allergic individuals. The high resistance of Pru p 3 to processing treatments and to digestion or enzymatic hydrolysis is probably the cause of the severity of this fruit allergy. The aim of this study was to determine the effect of treatment with a large number of proteases from different origins (vegetal, animal and microbial) on the degradation and allergenicity of Pru p 3.
