Understanding factors influencing the estimated genetic variance and the distribution of breeding values.

This study investigated the main factors influencing the genetic variance and the variance of breeding values (EBV). The first is the variance of genetic values in the base population, and the latter is the variance of genetic values in the population under evaluation. These variances are important as improper variances can lead to systematic bias. The inverse of the genetic relationship matrix ( ) and the phenotypic variance are the main factors influencing the genetic variance and heritability (h). These factors and h are also the main factors influencing the variance of EBVs. Pedigree- and genomic-based relationship matrices ( and as ) and phenotypes on 599 wheat lines were used. Also, data were simulated, and a hybrid (genomic-pedigree) relationship matrix ( as ) and phenotypes were used. First, matrix underwent a transformation (* = + ' + ), and the responses in the mean and variation of diag( ) and offdiag( ) elements, and genetic variance in the form of h were recorded. Then, the original was inverted, and matrix underwent the same transformations as , and the responses in the h estimate and the variance of EBVs in the forms of correlation and regression coefficients with the EBVs estimated based on the original were recorded. In response to weighting by , the estimated genetic variance changed by 1/. We found that (diag()) - (offdiag()) influences the genetic variance. As such, did not change the genetic variance, and increasing increased the estimated genetic variance. Weighting by was equivalent to weighting by 1/. Using the weighted together with its corresponding h, EBVs remained unchanged, which shows the importance of using variance components that are compatible with the . Increasing added to increased the estimated genetic variance, and the effect of ' was minor. We found that larger variation of diag( ) and higher concentration of offdiag( ) around the mean (0) are responsible for lower h estimate and variance of EBVs.

Astrocyte to neuron reprogramming has been performed using viral delivery of neurogenic transcription factors in GFAP expressing cells. Recent reports of off-target expression in cortical neurons following adeno-associated virus (AAV) transduction to deliver the neurogenic factors have confounded our understanding of the efficacy of direct cellular reprogramming. Genetic diversity can influence fitness components such as survival and reproductive success. Yet the association between genetic diversity and fitness based on neutral loci is sometime very weak and inconsistent, with relationships varying among taxa due to confounding effects of population demography and life history. Fitness-diversity relationships are likely to be stronger and more consistent for genes known to influence phenotypic traits, such as immunity-related genes, and may also depend on the genetic differences between breeding partners.
