• Cardiac rhabdomyomas are closely associated with tuberous sclerosis. • Obstructive CRs historically have required surgery. • Use of mTOR inhibitors can cause regression and involution of CRs. • Use of mTOR inhibitors can reduce morbidity/mortality associated with cardiac surgery.

Background: Abdominal aortic aneurysm (AAA) rupture is a significant cause of mortality in the elderly population. Despite experimental models identifying promising pharmacological therapies, there is still a lack of pharmacological interventions for AAA prior to surgery. This study aims to evaluate the regulatory role of the novel adenosine monophosphate-activated protein kinase (AMPK) agonist O304 in AAA formation and explore its underlying molecular mechanisms.

Cryo-EM Structure of AAA + ATPase Thorase Reveals Novel Helical Filament Formation.

bioRxiv

Neuroregeneration and Stem Cell Programs, Institute for Cell Engineering, The Johns Hopkins University School of Medicine, Baltimore, Maryland, United States of America.

The AAA+ (ATPases associated with a variety of cellular activities) ATPase, Thorase, also known as ATAD1, plays multiple roles in synaptic plasticity, mitochondrial quality control and mTOR signaling through disassembling protein complexes like AMPAR and mTORC1 in an ATP-dependent manner. The Oligomerization of Thorase is crucial for its disassembly and remodeling functions. We show that wild-type Thorase forms long helical filaments , dependent on ATP binding but not hydrolysis.

Objective: To investigate the mechanism by which Montelukast inhibits abdominal aortic aneurysm (AAA) formation through the AMPK/mTOR signaling pathway in mice.

Methods: Mice were randomly assigned to the Normal group, Model group, Montelukast group, and Montelukast + compound C (C.C) group.

Article Synopsis
  • - The study reviews current immunotherapy approaches for small bowel neuroendocrine tumors (SBNET) and explores future avenues for better treatment responses
  • - Key treatments like somatostatin analogs and mTOR inhibitors are essential, while new applications of peptide receptor radionuclide therapy (PRRT) show promise, though existing immunotherapies have had limited success
  • - Increased incidence of SBNET highlights the need for more effective therapies, and further research into the immune microenvironment may uncover new targets for improved patient outcomes

Aging is a gradual and irreversible physiological process that significantly increases the risks of developing a variety of pathologies, including neurodegenerative, cardiovascular, metabolic, musculoskeletal, and immune system diseases. Mitochondria are the energy-producing organelles, and their proper functioning is crucial for overall cellular health. Over time, mitochondrial function declines causing an increased release of harmful reactive oxygen species (ROS) and DNA, which leads to oxidative stress, inflammation and cellular damage, common features associated with various age-related pathologies.
