Regulatory T cells (Tregs) contribute significantly to the immunosuppressive nature of the tumor microenvironment which is a main barrier for immunotherapies of solid cancers. Reducing Treg numbers enhances anti-tumor immune responses but current depletion strategies also impair effector T cells (Teffs), potentially leading to reduced anti-tumor immunity and/or autoimmune diseases. CD137 has been identified as the most differentially expressed gene between peripheral Tregs and intratumoral Tregs in virtually all solid cancers.

Background: Folate receptor alpha (FRα) overexpression is seen in many cancers. Radioligand therapy (RLT) has emerged as a promising tool to target FRα and has been investigated previously, but further progression was limited due to high kidney retention and, subsequently, toxicity. To circumvent this, we present here the development of a [I]I-GMIB-conjugated anti-human FRα (hFRα) single-domain antibody (sdAb), with intrinsically fast renal clearance and concomitant low kidney retention.

Anti-Centromere Protein A Antibody Disrupts the Competence of Mouse Oocytes Matured In Vitro.

Am J Reprod Immunol

December 2024

Department of Obstetrics and Gynecology, Reproductive Medicine, Graduate School of Medicine, Chiba University, Chuo-ku, Chiba, Japan.

Introduction: Anticentromere autoantibodies are associated with refractory IVF/ET failure, but causality is unclear. Experimental models are needed.

Methods: Immature oocytes collected from 23-day-old mice were matured in vitro for 18 h in a culture medium containing an anti-human centromere protein A (CENP-A) polyclonal antibody, and those oocytes' maturity and chromosome/spindle structure were assessed.

Treatment Response of Donor Specific Antibodies and Forced Expiratory Volume in Lung Transplant Recipients With Antibody Mediated Rejection.

Transplant Proc

December 2024

University Health, University Health Transplant Institute, The University of Texas Health Science Center at San Antonio, San Antonio, Texas; The University of Texas at Austin, College of Pharmacy, Pharmacotherapy Division, Austin, Texas.

Article Synopsis
  • * A study of 15 lung transplant recipients showed that over half achieved significant reductions in DSAs, with 71% reaching stabilization in lung function (FEV1) six months post-treatment, despite some decline after peak recovery.
  • * The findings underscore the challenges of managing DSAs and the recovery of lung function, but also highlight the potential for stabilization in patients following AMR treatment.
