Dual-mode bioanalysis integrating photoelectrochemical (PEC) and other modes is emerging and allows signal cross-checking for more reliable results. Metal-organic frameworks (MOFs) have been shown to be attractive materials in various biological applications. This work presents the utilization of MOF encapsulation and stimuli-responsive decapsulation for dual-mode PEC and fluorescence (FL) bioanalysis. Photoactive dye methylene violet (MV) was encapsulated in zeolitic imidazolate framework-90 (ZIF-90) to form an MV@ZIF-90 hybrid material, and MV could be released by adenosine triphosphate (ATP)-induced ZIF-90 disintegration. The released MV not only had FL emission but also had a sensitization effect on the ZnInS (ZnInS) photoanode. Based on the MV-dependent sensitization effect and FL emission characteristic, a dual-mode PEC-FL strategy was established for ATP detection with low detection limits, that is, 3.2 and 4.1 pM for PEC and FL detection, respectively. This study features and will inspire the construction and implementation of smart MOF materials for dual-mode bioanalysis.

Accurate and sensitive detection of Pax-5a gene is the basis of early diagnosis and prediction of acute leukemia. This research aims to develop a universal dual-mode sensing method enables ultrasensitive gene detection based on smart control of DNA amplification by nucleic acid beacons e to form programmed dendrimer. The Pax-5a target gene triggers the opening of smart gate hairpin probe (Hp), exposing the stem sequence as the primer to bind with padlock probe for rolling circle amplification (RCA).

To advance the biological understanding of heat shock protein (HSP) in different types of cancers, it is crucial to achieve its accurate determination. Herein, a dual-mode self-powered photoelectrochemical (PEC) and colorimetric platform was proposed by integrating enzymatic catalysis and a chemical redox cycling amplification strategy. In this system, ascorbic acid (AA), as the signal reporter for PEC and colorimetric assay, can be regenerated during the tris(2-carboxyethyl) phosphine-mediated chemical redox cycling process.

Dual mode detection can overcome the poor anti-interference ability of single-mode detection, and greatly improve the detection accuracy. Fluorescence/electrochemiluminescence (FL/ECL) dual mode detection combines the advantages of FL and ECL, and has a promising application in bioanalysis. Common FL/ECL dual mode detection used different signal probes.

Background: Bisphenol A (BPA) has been identified as an endocrine disruptor with numerous detrimental effects on human health. There is an urgent need to develop fluorescence/colorimetric dual-mode sensing approaches with expanded detection linear range, increased accuracy, and enhanced application flexibility for BPA detection. The utilization of fluorescence and colorimetric signals in point-of-care applications and real-time sensitive sensing further highlights the significance of developing novel and efficient fluorescence/colorimetric dual-mode sensing platform with high-efficiency probes.
