The hippocampus is involved in processing a variety of mnemonic computations specifically the spatiotemporal components and emotional dimensions of contextual memory. Recent studies have demonstrated cellular heterogeneity along the hippocampal axis. The ventral hippocampus has been shown to be important in the processing of emotion and valence. Here, we combine transgenic and all-virus based activity-dependent tagging strategies to visualize multiple valence-specific engrams in the vHPC and demonstrate two partially segregated cell populations and projections that respond to appetitive and aversive experiences. Next, using RNA sequencing and DNA methylation sequencing approaches, we find that vHPC appetitive and aversive engram cells display different transcriptional programs and DNA methylation landscapes compared to a neutral engram population. Additionally, optogenetic manipulation of tagged cell bodies in vHPC is not sufficient to drive appetitive or aversive behavior in real-time place preference, stimulation of tagged vHPC terminals projecting to the amygdala and nucleus accumbens (NAc), but not the prefrontal cortex (PFC), showed the capacity drive preference and avoidance. These terminals also were able to change their capacity to drive behavior. We conclude that the vHPC contains genetically, cellularly, and behaviorally segregated populations of cells processing appetitive and aversive memory engrams.

Affective processing is important for guiding behavior and its dysfunction can lead to several psychiatric illnesses, including depression and substance use disorders. Conditioned taste aversion (CTA) is used to study learned shifts in affect, and taste reactivity (TR) can effectively track the hedonic properties of appetitive and aversive tastants before and after CTA. While the infralimbic cortex (IL) and its projections to the nucleus accumbens (NAc) shell play a key role in learned negative affect, this role is unique to males.

The personality trait of neuroticism has been theoretically linked to threat sensitivity, but this perspective of neuroticism has resulted in mixed findings, arguably because mood states, rather than emotional reactions, have been examined. The present studies (total = 519) administered a task capable of assessing emotional reactions-to appetitive versus aversive images-in a nearly continuous manner, parsing threat sensitivity in terms of emotional onsets, peak amplitudes, and prototypicality in responding. In the context of this tight temporal focus, higher levels of neuroticism tended to be associated with faster emotional onsets when aversive images were involved.

Ventral tegmental area (VTA) dopamine (DA) neurons are classically linked to Pavlovian reward learning and reinforcement. Intermingled VTA GABA neurons are positioned to regulate dopaminergic and striatal systems, but we lack critical insight into how this population contributes to conditioned motivation in different learning contexts. Recording DA and GABA neurons across multiple conditioning paradigms, we found that GABA neurons not only actively encode appetitive and aversive cues and outcomes separately, but uniquely integrate salient events of both valences to guide reward seeking.
