Reproduction in females is an energetically demanding process. We assumed that adiponectin (ADPN), known for its role in energy balance maintenance, is also engaged in the regulation of uterine steroidogenesis in the pig. We determined the impact of ADPN alone or in combination with insulin (INS) on testosterone (T), estrone (E) and estradiol (E) secretion by porcine endometrium and myometrium, uterine expression of CYP17A1 and CYP19A3 genes, and endometrial abundance of P450 and P450 proteins during the peri-implantation period and the oestrous cycle, using radioimmunoassay, qPCR, and Western Blot, respectively. During pregnancy, in the endometrial explants from days 10-11, ADPN decreased CYP17A1 gene expression, P450 protein abundance and T secretion, whereas increased E secretion. On days 12-13 of pregnancy, ADPN decreased CYP17A1 and CYP19A3 expression, P450 and P450 protein abundance and E secretion, but stimulated T secretion. On days 15-16 of pregnancy, ADPN decreased P450 protein accumulation but enhanced CYP19A3 expression and E secretion. On days 27-28 of pregnancy, ADPN increased CYP17A1 and CYP19A3 mRNA content and T secretion in this tissue and decreased P450 content. ADPN effect on myometrial explants was dependent on stage of gestation or oestrous cycle. Moreover, INS treatment modulated basal and ADPN-affected steroidogenic enzymes gene and protein expression and steroids secretion. The results obtained indicate that ADPN may affect processes required for successful implantation such as steroidogenesis. ADPN and INS were also shown to modulate each other action, which indicates that the proper course of uterine steroidogenesis may be dependent on both hormones' interaction.

Role of resistin in the porcine uterus: effects on endometrial steroidogenesis.

Reprod Fertil Dev

January 2025

Department of Animal Anatomy and Physiology, Faculty of Biology and Biotechnology, University of Warmia and Mazury in Olsztyn, Olsztyn, Poland.

Context The adipose tissue produces adipokines - hormones essential to many biological functions, including reproduction. Aims We hypothesised that resistin, one of the adipokines, is present in the blood plasma, uterine luminal flushings (ULF) and uterus of pigs during the oestrous cycle and early pregnancy, and that resistin influences uterine steroidogenesis. Methods This study aimed to determine the expression of resistin in the porcine endometrium and myometrium during the cycle and pregnancy by quantitative real-time polymerase chain reaction and western blot (WB).

Unlabelled: The present study examined the anti-inflammatory and functional improvement of the uterus and ovary, respectively, in bisphenol-A (BPA)-fed adult Wistar rats following the ingestion of methanolic extract of wheatgrass (WG-ME). Four groups of rats were conditioned as vehicle-treated control, BPA-treated (100 mg/kg b.w.

Article Synopsis
  • * Most existing studies focus on single phthalate exposures in animal models, leaving the effects of real-life mixtures and their impacts on women's reproductive health largely unexamined.
  • * Recent literature indicates that high phthalate exposure is linked to menstrual irregularities, ovarian disorders, poor IVF outcomes, and adverse pregnancy results, highlighting the need for better research on phthalate mixtures and their mechanisms of toxicity.

To date five members of estrogen receptors (ESRs) have been reported. They are grouped into two classes, the nuclear estrogen receptors are members of the nuclear receptor family which found at nuclear, cytoplasm and plasma membrane, and the membrane estrogen receptors, such as G protein-coupled estrogen receptor 1, ESR-X and Gq-coupled membrane estrogen receptor. The structure and function of estrogen receptors, and interaction between ESR and coregulators were reviewed.

Article Synopsis
  • Cumulus granulosa cells (CGCs) are critical for oocyte growth and reproduction in mammals, making it important to study their molecular functions.
  • This research focused on the yak MSX2 gene, showing it encodes a protein that is highly expressed in female reproductive organs, particularly during the luteal phase of the estrous cycle.
  • Manipulating MSX2 levels in yak CGCs revealed its impact on cell viability and steroid hormone production, providing new insights into yak reproductive mechanisms.
