Melanin biosynthesis is enzymatically regulated by tyrosinase (TYR, EC 1.14.18.1), which is efficiently inhibited by natural and synthetic phenols, demonstrating potential therapeutic application for the treatment of several human diseases. Herein we report the inhibitory effects of a series of (4-(4-hydroxyphenyl)piperazin-1-yl)arylmethanone derivatives, that were designed, synthesised and assayed against TYR from Agaricus bisporus (AbTYR). The best inhibitory activity was predominantly found for compounds bearing selected hydrophobic ortho-substituents on the aroyl moiety (IC values in the range of 1.5-4.6 μM). They proved to be more potent than the reference compound kojic acid (IC =17.8 μM) and displayed competitive mechanism of inhibition of diphenolase activity of AbTYR. Docking simulation predicted their binding mode into the catalytic cavities of AbTYR and the modelled human TYR. In addition, these compounds displayed antioxidant activity combined with no cytotoxicity in MTT tests. Notably, the best inhibitor affected tyrosinase activity in α-MSH-stimulated B16F10 cells, thus demonstrating anti-melanogenic activity.

Pyrogenic carbons (PCs), with varying structures depending on the materials and thermal treatment conditions, have been extensively used to enhance anaerobic digestion by mediating electron transfer. However, the underlying mechanism has yet to be explored. Herein, the redirection and enhancement of the direct interspecies electron transfer (DIET) pathway were evidenced, along with the upregulated electrochemical properties and structural proteins in the methanogenic consortia.

Aims: This study aimed to evaluate the impact of community-based dental education (CBDE) on the learning experiences of undergraduate dental students and recent dental graduates from two diverse geographical regions.

Methods: The study followed a cross-sectional design, conducted online using Google Forms, with ethical approval from Qatar University. A non-probability purposive sampling method was used to recruit dental students and recent graduates from three institutions in India and one in Qatar.

Rac/Rop proteins, a kind of unique small GTPases in plants, play crucial roles in plant growth and development and in response to abiotic and biotic stresses. However, it is poorly understood whether cotton Rac/Rop protein genes are involved in mediating cotton resistance to Verticillium dahliae. Here, we focused on the function and mechanism of cotton Rac/Rop gene GhRac9 in the defense response to Verticillium dahliae infection.

Quantification of ivermectin in veterinary products consumed off-label as a treatment for COVID-19.

Centre of Excellence for Pharmaceutical Sciences (Pharmacen™), North-West University, Potchefstroom, Republic of South Africa.

The COVID-19 pandemic caused global pandemonium, and due to an unprecedented global response, the popularity and use of (veterinary) ivermectin, amongst many other conceivable 'treatments', experienced a meteoric rise. Ivermectin is a macrocyclic lactone compound belonging to the avermectin drug class and is a registered medicine in many countries, although the most common use is as veterinary medicine. In this study, a fast HPLC method was developed and validated for the quantification of ivermectin in veterinary products that were used off-label by a substantial number of people during COVID-19.

Cigarette smoke extract (CSE)-induced airway mucus hypersecretion and inflammation are prominent features of chronic obstructive pulmonary disease (COPD). As a factor associated with inflammation regulation, T cell immunoglobulin and mucin domain-1 (TIM-1) is found to be involved in various inflammatory disorders such as asthma and COPD. In this study, the GEO database provides two human COPD gene expression datasets (GSE67472, n = 62) along with the relevant controls (n = 43) for differentially expressed gene (DEG) analyses.
