Targeting MAPK14 by Lobeline Upregulates Slurp1-Mediated Inhibition of Alternative Activation of TAM and Retards Colorectal Cancer Growth.

Adv Sci (Weinh)

January 2025

Jiangsu Collaborative Innovation Center of Traditional Chinese Medicine in Prevention and Treatment of Tumor, The First Clinical College, Nanjing University of Chinese Medicine, 138 Xianlin Avenue, Nanjing, 210023, China.

Colorectal cancer (CRC) usually creates an immunosuppressive microenvironment, thereby hindering immunotherapy response. Effective treatment options remain elusive. Using scRNA-seq analysis in a tumor-bearing murine model, it is found that lobeline, an alkaloid from the herbal medicine lobelia, promotes polarization of tumor-associated macrophages (TAMs) toward M1-like TAMs while inhibiting their polarization toward M2-like TAMs.

Masticatory muscle changes on magnetic resonance imaging of dogs with compared to meningoencephalitis of unknown origin.

Front Vet Sci

January 2025

Anderson Moores Veterinary Specialists, Linnaeus Veterinary Limited, Winchester, United Kingdom.

Infectious meningoencephalitides represent an important differential diagnosis for meningoencephalitis of unknown origin (MUO) in dogs. Treatment of the latter requires immunosuppression, but laboratory test results for infectious agents may take several days to return. This study investigated whether the presence of masticatory muscle changes on magnetic resonance imaging (MRI) of the head can be used to distinguish dogs with neosporosis from those with MUO at the time of diagnosis.

Fournier's gangrene (FG) is a type of necrotizing fasciitis affecting the abdomen or perineum. It is a polymicrobial infection that progresses to an obliterating endarteritis, causing thrombosis and subsequent tissue necrosis, allowing pathogenic invasion of interfacial planes.Patients with Fournier's gangrene typically have underlying systemic conditions that cause vascular insufficiencies or immunosuppression.

Background: Paroxysmal nocturnal hemoglobinuria (PNH) is a rare complement-driven acquired hemolytic anemia with specific presentations of hemoglobinuria, abdominal pain, fatigue, and thrombosis.

Objective: To review the current therapeutic strategies for PNH, including anti-complement therapy and allogeneic hematopoietic cell transplantation (alloHCT), focusing on the tailoring of the approach to the disease subtype.

Results: The outcome of alloHCT varies depending on disease severity, thrombotic history, and response to prior therapies.

Transplant oncology and anti-cancer immunosuppressants.

Front Immunol

January 2025

Tianjin Organ Transplantation Research Center, Tianjin First Central Hospital, Nankai University School of Medicine, Tianjin, China.

Organ transplantation is a life-saving intervention that enhances the quality of life for patients with end-stage organ failure. However, long-term immunosuppressive therapy is required to prevent allogeneic graft rejection, which inadvertently elevates the risk of post-transplant malignancies, especially for liver transplant recipients with a prior history of liver cancer. In response, the emerging field of transplant oncology integrates principles from oncology and immunology to improve outcomes for patients at high risk of tumor occurrence or recurrence following transplantation.
