Phosphoinositide 3-kinase (PI3K), a member of the class I PI3K family, is an essential signaling biomolecule that regulates the differentiation, proliferation, migration, and survival of immune cells. The overactivity of this protein causes cellular dysfunctions in many human disorders, for example, inflammatory and autoimmune diseases, including asthma or chronic obstructive pulmonary disease (COPD). In this work, we designed and synthesized a new library of small-molecule inhibitors based on indol-4-yl-pyrazolo[1,5-]pyrimidine with IC values in the low nanomolar range and high selectivity against the PI3K isoform. CPL302253 (), the most potent compound of all the structures obtained, with IC = 2.8 nM, is a potential future candidate for clinical development as an inhaled drug to prevent asthma.

The therapeutic benefits of opioids are compromised by the development of analgesic tolerance, which necessitates higher dosing for pain management thereby increasing the liability for drug dependence and addiction. Rodent models indicate opposing roles of the gut microbiota in tolerance: morphine-induced gut dysbiosis exacerbates tolerance, whereas probiotics ameliorate tolerance. Not all individuals develop tolerance, which could be influenced by differences in microbiota, and yet no study design has capitalized upon this natural variation.

Background: Recent studies indicate that N6-methyladenosine (mA) RNA modification may regulate ferroptosis in cancer cells, while its molecular mechanisms require further investigation.
Methods: Liquid Chromatography-Tandem Mass Spectrometry (HPLC/MS/MS) was used to detect changes in mA levels in cells. Transmission electron microscopy and flow cytometry were used to detect mitochondrial reactive oxygen species (ROS).

Background: Tissue metabolomics analysis, alongside genomics and proteomics, offers crucial insights into the regulatory mechanisms of tumorigenesis. To enhance metabolite detection sensitivity, chemical isotope labeling (CIL) techniques, such as dansylation, have been developed to improve metabolite separation and ionization in mass spectrometry (MS). However, the dissolution of hydrophobic derivatized metabolites in solvents with high acetonitrile content limits the use of liquid chromatography (LC) systems with small-volume reversed-phase (RP) columns.

Hypochlorous acid (HClO) is a well-known inflammatory signaling molecule, while lipid droplets (LDs) are dynamic organelles closely related to inflammation. Using organic small-molecule fluorescence imaging technology to target LDs for precise monitoring of HClO is one of the most effective methods for diagnosing inflammation-related diseases. A thorough investigation of how probes detect biological markers and the influencing factors can aid in the design of probe molecules, the selection of high-performance tools, and the accuracy of disease detection.

Background: Carbosulfan residues in environment is very harmful to human health. The rapid and high sensitive detection of carbosulfan residues is particularly important to guarantee human health and safety. The conventional chromatographic techniques and enzyme inhibition strategies cannot realize on-site and visual detection of carbosulfan.
