Although multivalent glucosidase inhibitors based on iminosugars have shown enhanced inhibition activity, an effective way to improve their hypoglycemic effect in vivo, is still in infancy and needs further development. In this paper, PBI-5DNJ and PBI-6DNJ, with three or four DNJ moieties respectively conjugated at the bay position were synthesized. PBI-6DNJ evidenced stronger π-π stacking interactions and, when self-assembled, a smaller size than that of PBI-5DNJ. It was found that PBI-6DNJ exhibited superior α-glucosidases (from mice) inhibition activity (K = 0.14 ± 0.007 μM) in vitro than that (K = 0.31 ± 0.01) of PBI-5DNJ and in vivo hypoglycemic effects in mice models. PBI-6DNJ possessed good hypoglycemic effects with the percentages of PBG levels of 40.40 ± 3.33% and 39.23 ± 4.84% at a dose of 2.0 mg/kg after 15 min and 30 min of administration, respectively. In terms of measuring percentage decrease of PBG level per DNJ unit, PBI-6DNJ displayed a 2.1-fold enhancement than miglitol, demonstrating a consistency between in vitro and in vivo experiments. This paves the way to the connection between in vivo hypoglycemic potency and in vitro glucosidase inhibition assay, leading to reliable and simplified assessment of hypoglycemic potency determination, and opening a basic understanding of the design of multivalent glucosidase inhibitors.

A concise asymmetric synthesis of clickable enantiomeric pyrrolidines was achieved using Crabbé-Ma allenation. The synthesized iminosugars were grafted by copper-free strain-promoted alkyne-azide cycloaddition onto phosphorus dendrimers. The hexavalent and dodecavalent pyrrolidines were evaluated as β-glucocerebrosidase inhibitors.

Article Synopsis
  • - Researchers are developing multivalent glycosidase inhibitors derived from 1-deoxynojirimycin to target α-glucosidases, but the effectiveness of self-assembly in living systems still needs more investigation.
  • - They created two amphiphilic compounds and examined their ability to self-assemble, inhibit α-glucosidases, and provide fluorescence imaging in living organs.
  • - One compound showed better inhibition and successfully formed a complex that could be visualized in the small intestine, suggesting that this self-assembly strategy could lead to new hypoglycemic drug designs.

The first phosphorus dendrimers built on a cyclotriphosphazene core and decorated with six or twelve monofluorocyclooctyne units were prepared. A simple stirring allowed the grafting of N-hexyl deoxynojirimycin inhitopes onto their surface by copper-free strain promoted alkyne-azide cycloaddition click reaction. The synthesized iminosugars clusters were tested as multivalent inhibitors of the biologically relevant enzymes β-glucocerebrosidase and acid α-glucosidase, involved in Gaucher and Pompe lysosomal storage diseases, respectively.

Article Synopsis
  • Synthetic glycoconjugates are being developed to detect glycosidase enzymes, but effective binding interactions, particularly with multivalent glycosidase inhibitors and α-glycosidases, remain limited.
  • The study synthesizes three naphthalimide-DNJ conjugates and investigates their binding and inhibition effects on glycosidases, finding that their inhibitory activity is linked to strong binding interactions.
  • Among the compounds, compound 13, which features a six-carbon chain, demonstrated the strongest binding and inhibition against specific glycosidases, resulting in a significant reduction in blood glucose levels in mice, suggesting potential for fluorescent sensing methods in detecting glycosidase inhibition.
