A Pd-catalyzed alkyne and aryne annulation strategy via C-H activation has been implemented for the synthesis of πextended coumarins. This synthetic strategy provides a wide range of πextended coumarins in moderate to good yields with good functional group compatibility. Photophysical properties of the synthesized π-extended coumarins have been evaluated, and some of them show interesting fluorescent properties. Three of the synthesized coumarins have been unambiguously established by a single-crystal XRD study.

ConspectusControlling selectivity through manipulation of reaction intermediates remains one of the most enduring challenges in organic chemistry, providing novel solutions for selective C-C π-bond functionalization. This approach, guided by activation principles, provides an effective method for selective functional group installation, enabling direct synthesis of organic molecules that are inaccessible through conventional pathways. In particular, the selective functionalization of N-conjugated allenes and alkynes has emerged as a promising research focus, driven by advances in controlling reactive intermediates and activation strategies.

An unprecedented Pd-catalyzed cascade alkyne insertion/Heck/C-H activation reaction of -iodophenyl alkenyl ethers and diarylacetylenes has been developed. Diversified tetracyclic-fused dihydroindeno[2,1-]chromenes bearing a quaternary center were constructed in an efficient, straightforward, and atom-economic way with good to excellent yields. The protocol features high bonding efficiency, operational simplicity, broad substrate scope, and easy scale-up.

A novel and highly efficient Pd-catalyzed approach for the synthesis of bis-heterocycles featuring both isoxazoline and methyleneindole motifs is demonstrated. The in situ formation of vinyl Pd(II) species through an alkyne-tethered carbamoyl chloride cyclization is crucial, and the innovative Pd-catalyzed carboetherification of β,γ-unsaturated oximes with vinyl Pd(II) species has been developed. This method is not only operationally straightforward but also exhibits a broad substrate scope and excellent functional group tolerance.

Pd-catalyzed alkoxycarbonylation of internal alkynes provides a straightforward access to α,β-disubstituted acrylic esters. Compared with the well-established regioselective alkoxycarbonylation of terminal alkynes, the regioselective hydrocarboxylation of non-functionalized unsymmetric internal alkynes was more challenging owing to the delicate differences of properties between the two substituents. Herein, by using either monophosphine ligand based on 2,3-dihydrobenzo[d][1,3]oxaphosphole motif or bidentate ligand Ph-Phox, the regioselective alkoxycarbonylations of aryl-aryl, aryl-alkyl and alkyl-alkyl disubstituted alkynes were achieved, giving a diversity of trisubstituted α,β-unsaturated carboxylic esters with moderate to excellent yields and high regioselectivity.
