To investigate heterogeneity of fibroblasts in human fetal skin, we analysed published single-cell RNA sequencing data (8 and 16 post conception weeks (PCW)) and performed single-molecule fluorescence in situ hybridisation to map their spatial distribution and predicted dynamic interactions. Clustering revealed 8 fibroblast populations with developmental stage-specific abundance changes. Proliferative cells (MKI67+) were present at all stages.

Hoffmann syndrome, a rare manifestation of hypothyroid myopathy in adults, is characterised by muscle weakness, stiffness and pseudohypertrophy. Here, we report the case of a middle-aged man who presented with progressive weakness in proximal muscles (in the form of difficulty in climbing stairs, rising from a seated position, combing hair and lifting objects) and leg swelling for 6 months. Physical examination revealed pseudohypertrophy of calf muscles with pronounced symmetric weakness in proximal upper and lower limbs.

Although the interconnection between dysregulated stress systems and FMS is well documented, the interconnection between early adversities and FMS remains less understood. This study explores the relationship of early-life stress and FMS by examining its mediation through perceived stress, and acute and chronic endocrine stress indicators.
