To ensure the correct euploid state of embryos, it is essential that vertebrate oocytes await fertilization arrested at metaphase of meiosis II. This MII arrest is mediated by XErp1/Emi2, which inhibits the ubiquitin ligase APC/C (anaphase-promoting complex/cyclosome). Cyclin B3 in complex with Cdk1 (cyclin-dependent kinase 1) is essential to prevent an untimely arrest of vertebrate oocytes in meiosis I by targeting XErp1/Emi2 for degradation.

Article Synopsis
  • Secretory proteins rely on the signal peptidase complex (SPC) for proper processing of their signal sequences, which is vital for correct protein folding and placement in eukaryotic cells.
  • The study focused on the role of Spc2 within the SPC, showing that mutations or depletion of Spc2 hinder the complex’s ability to recognize and cleave substrates correctly.
  • Molecular dynamics simulations revealed that without Spc2, the SPC's membrane structure is altered, shedding light on how Spc2 affects the protein biogenesis process.

Chromosome alignment on the metaphase plate is a conserved phenomenon and is an essential function for correct chromosome segregation for many organisms. Organisms with naturally-occurring trivalent chromosomes provide a useful system for understanding how chromosome alignment is evolutionarily regulated, as they align on the spindle with one kinetochore facing one pole and two facing the opposite pole. We studied chromosome alignment in a praying mantid that has not been previously studied chromosomally, the giant shield mantis .

Unlabelled: Membrane trafficking is a conserved process required for the movement and distribution of proteins and other macromolecules within cells. The NIMA-related kinases NEKL-2 (human NEK8/9) and NEKL-3 (human NEK6/7) are conserved regulators of membrane trafficking and are required for the completion of molting. We used a genetic approach to identify reduction-of-function mutations in that suppress -associated molting defects.

Chronic stress-related brain disorders are widespread and debilitating, and often cause lasting neurobehavioral deficits. Minocycline, a common antibiotic and an established inhibitor of microglia, emerges as potential treatment of these disorders. The zebrafish (Danio rerio) is an important emerging model organism in translational neuroscience and stress research.
