Polyampholyte nanogels based on -isopropylacrylamide (NIPAM), (3-acrylamidopropyl) trimethylammonium chloride (APTAC) and 2-acrylamido-2-methyl-1-propanesulfonic acid sodium salt (AMPS) were synthesized via conventional redox-initiated free radical copolymerization. The resultant nanogels of various compositions, specifically [NIPAM]:[APTAC]:[AMPS] = 90:5:5; 90:7.5:2.5; 90:2.5:7.5 mol.%, herein abbreviated as NIPAM-APTAC-AMPS, NIPAM-APTAC-AMPS and NIPAM-APTAC-AMPS, were characterized by a combination of H NMR and FTIR spectroscopy, TGA, UV-Vis, DLS and zeta potential measurements. The temperature and salt-responsive properties of amphoteric nanogels were studied in aqueous and saline solutions in a temperature range from 25 to 60 °C and at ionic strengths (μ) of 10 to 1M NaCl. Volume phase transition temperatures (VPTT) of the charge-balanced nanogel were found to reach a maximum upon the addition of salt, whereas the same parameter for the charge-imbalanced nanogels exhibited a sharp decrease at higher saline concentrations. A wide bimodal distribution of average hydrodynamic sizes of nanogel particles had a tendency to transform to a narrow monomodal peak at elevated temperatures and higher ionic strengths. According to the DLS results, increasing ionic strength results in the clumping of nanogel particles.

Polyamidoamines (PAAs) are a well-known family of synthetic biocompatible and biodegradable polymers, which can be prepared as soft hydrogels characterized by low interfacial tension and tunable elasticity. For the first time we report here on the in vivo performance of a PAA hydrogel implant as scaffold for tissue engineering. In particular, an amphoteric agmatine-deriving PAA hydrogel shaped as small tubing was obtained by radical polymerization of a soluble functional oligomeric precursor and used as conduit for nerve regeneration in a rat sciatic nerve cut model.

Electrical behavior of a natural polyelectrolyte hydrogel: chitosan/carboxymethylcellulose hydrogel.

Biomacromolecules

April 2008

The Key Laboratory of Molecular Engineering of Polymers of MOE, Department of Macromolecular Science, Advanced Material Laboratory, Fudan University, Shanghai, 200433, People's Republic of China.

An amphoteric hydrogel film was prepared by solution blending of two natural polyelectrolytes, chitosan and carboxymethylcellulose, and cross-linking with glutaraldehyde. The bending of the film in an electric field was studied in different electrolyte solutions. Because of its amphoteric nature, the hydrogel can bend toward either anode or cathode depending on the pH of the solution.

The geometric characteristics of nanogel particles in aqueous solutions were studied by determining their ratios of radius of gyration (mean-square radius; Rg) to hydrodynamic radius (Rh), Rg/Rh, derived from static light scattering and dynamic light scattering experiments, respectively. The various nanogel samples studied included ones composed of lightly cross-linked N-isopropylacrylamide (NIPA) polymer, NIPA-based anionic or cationic copolymers, and amphoteric terpolymers. Polyelectrolyte complexes between anionic or cationic nanogels and oppositely charged polyions or nanogels having opposite charges were also studied.
