Two new pyranone derivatives phomapyrone A (2) and phomapyrone B (3), one new coumarin 11 13-(+)-phomacumarin A (1), three known pyranones (4-6), together with three known amide alkaloids fuscoatramides A-C (7-9), as well as 9 11-(+)-ascosalitoxin (10) were isolated from the endophytic fungus sp. YN02-P-3, which was isolated from the healthy leaf tissue of a Paulownia tree in Yunnan Province, China. Their structures were elucidated using extensive NMR spectroscopic and HRESIMS data and by comparing the information with literature data. In addition, all compounds were tested for their cytotoxicity activity against human tumor cell lines, and the results showed that new compounds 1-3 showed moderate inhibitory activity against the HL-60 cell line with IC values of 31.02, 34.62, and 27.90 M, respectively.

In this computational study, density functional theory (DFT) and time-dependent DFT methods (TD-DFT) were employed to study the optical properties of six families of molecules with donor (D), bridge (B), and acceptor (A) fragments that have potential for use as fluorescent molecular probes for the early detection of Alzheimer's disease. After validating our computational method against experimental data, using X-ray and absorption data, the equilibrium geometries and wave functions of the ground and first singlet excited states were systematically studied. Our simulations demonstrate that the S states of these rod-like D-B-A fluorescent probes are twisted intramolecular charge transfer states with a predominant highest occupied molecular orbital-least unoccupied molecular orbital (HOMO-LUMO) character, the former localized primarily at the donor, whereas the latter at the acceptor site.

For the first time, the interaction of aroyl containing pyrano[2,3-]isoxazolone derivatives with various hydrazines was studied. It was shown that the considered process includes formation of corresponding hydrazones followed by Boulton-Katritzky rearrangement. As a result, the general method for the synthesis of substituted 1,2,3-triazoles bearing an allomaltol fragment was elaborated.
