The present protocol describes a bioluminescence reporter assay developed to quantify the ability of synthetic agonists of retinoic acid receptors (RARs) to activate glutamate receptor subunit 1 (GluR1) translation. The reporter assay uses firefly luciferase under the control of the GluR1 5' untranslated region (5' UTR) which is bound by RARs to regulate its translation. This method is used to demonstrate the role of RARα in retinoic acid regulation of GluR1 translation. This method may also be used to screen drugs that influence RAR induction of GluR1 translation as an important mechanism controlling learning and memory in the brain.

Purpose: To investigate BMAL1 and BMP6 expressive differences in ovarian granulosa cells (OGCs) of patients with polycystic ovary syndrome (PCOS), explore regulatory relationship, assess their impacts on OGC proliferation and apoptosis, and analyze their correlations with ART outcomes of patients.
Methods: A clinical study selected 40 PCOS patients who underwent IVF/ICSI in our hospital from January to October 2022 and 39 controls with male or tubal factor infertility. RT-qPCR and Western blot assessed BMAL1 and BMP6 mRNA/protein levels.

Firefly luciferases have been extensively used for bioanalytical applications, including their use as bioluminescent reporters, biosensors, and for bioimaging biological and pathological processes. Due to their intrinsic pH- sensitivity, in recent years we have demonstrated that firefly luciferases can also be harnessed as color- tuning sensors of intracellular pH. However, it is known that mammalian cells require temperatures higher than 36 °C, which red-shift the bioluminescence spectra of most firefly luciferases, decreasing their activities and the resolution of ratiometric pH analysis.
