AI Article Synopsis

  • Microtaggant technologies encode unique numbers on pharmaceutical coatings to help authenticate products and fight counterfeit medicines.
  • The study introduced a new microtaggant called the stealth nanobeacon (NB), which uses surface-enhanced Raman scattering (SERS) for improved stability on tablet coatings.
  • Testing showed that while some SERS signal intensity declined under stress conditions, the NB maintained an adequate signal for effective authentication across various coating types.

Article Abstract

Microtaggant technologies can encode individual numbers on coating tablet to authenticate pharmaceutical products and, therefore, combat the global spread of falsified medicine. In this study, a novel microtaggant, stealth nanobeacon (NB), with surface-enhanced Raman scattering (SERS) activity was applied to various coating tablets and its physical stability was evaluated. The NBs were composed of a reporter molecule (AH, adenine hydrochloride) and prepared with different sizes of gold nanoparticles (AuNPs). The NBs were directly deposited on the surface of various model coatings (e.g., hydroxypropyl cellulose, hydroxypropyl methyl cellulose, Eudragit® RS30D, ethyl cellulose). To investigate physical stability of the NB on the coating tablets, SERS spectra of the NB after friability test and acceleration test (store at 75% RH, 40 °C) were evaluated using a portable Raman spectrometer. After the friability test, there was no significant decrease in the peak intensity of the SERS signal (P) for authentication in all samples. In the acceleration test, the SERS signals of the samples were attenuated, but sufficient SERS signal intensity (P > 70) was maintained in the seven types of coating for authentication. These results demonstrate that the microtaggant NB has the potential to be used for a wide range of coating tablets.

Film-coating is a critical step in pharmaceutical manufacturing. Traditional visual inspections for film-coated tablet defect assessment are subjective, inefficient, and labor-intensive. We propose a novel approach utilizing machine learning and image analysis to address these limitations.

Orodispersible film (ODF) is one of the novel formulations that disintegrate rapidly in the mouth without the requisite for water compared to other conventional oral solid dosage formulations. This delivery system serves as a convenient mode of administration, especially in patients who have dysphagia and fluid restriction, being beneficial to pediatric, geriatric, and bedridden patients. A novel sildenafil ODF containing sildenafil citrate is formulated to be used in patients with erectile dysfunction (ED).

Establishment of a suitable animal model to evaluate sustained release (SR) formulations is very important because it reduces the development time of SR formulations. Beagle dogs are often used to evaluate prototype formulations since they can be directly administered powder, such as drug substance. However, the physiological condition of dogs is very different to that of humans.

Utilising terahertz pulsed imaging to analyse the anhydrous-to-hydrate transformation of excipients during immediate release film coating hydration.

Pharmaceutical tablets are routinely film-coated to improve appearance, reduce medication errors and enhance storage stability. Terahertz pulsed imaging (TPI) can be utilised to study the liquid penetration into the porous tablet matrix in real time. Using polymer-coated flat-faced tablets with anhydrous lactose or mannitol, we show that when the tablet matrix contains anhydrous material, the anhydrous form transforms to the solid-state hydrate form in the tablet core while the immediate release coating dissolves.
