Macromolecular cryoprotectants based on polyampholytes are showing promise as supplemental cryoprotectants alongside conventional DMSO-based freezing. Here we exploit radical ring-opening (ter)polymerization to access ester-containing cryoprotective polyampholytes, which were shown to be degradable. Using a challenging cell monolayer cryopreservation model, the degradable polyampholytes were found to enhance post-thaw recovery when supplemented into DMSO. This demonstrates that degradable macromolecular cryoprotectants can be developed for application in biotechnology and biomedicine.

This study reports the fabrication of non-woven fibrous membranes from electrospinning blended solutions of PVDF with polyampholytes in -dimethylformamide and methanol. Polyampholytes are macromolecules that have both positive and negative charged units in different side groups attached to the backbone. In this study, we used a random polyampholyte amphiphilic copolymer (r-PAC) synthesized by co-polymerizing a hydrophobic monomer in addition to the positive and negative charged monomer units, to reduce the fouling propensity of PVDF electrospun membranes while preserving its inherent hydrophobicity.

Despite continuous expansion of the RNA binding protein (RBP) world, there is a lack of systematic understanding of RBPs in the mammalian testis, which harbors one of the most complex tissue transcriptomes. We adapted RNA interactome capture to mouse male germ cells, building an RBP atlas characterized by multiple layers of dynamics along spermatogenesis. Trapping of RNA-cross-linked peptides showed that the glutamic acid-arginine (ER) patch, a residue-coevolved polyampholytic element present in coiled coils, enhances RNA binding of its host RBPs.

Over last years, hydrogels based on natural polymers have attracted considerable interest as materials for wound healing. Herein, hydrogel films based on kappa-carrageenan and guanidinium polyampholytes were prepared by the in situ physical cross-linking with potassium chloride and borax, respectively. Tuftelin Interacting Protein 11 (TFIP11) was identified as a critical human spliceosome assembly regulator, interacting with multiple proteins and localising in membrane-less organelles. However, a lack of structural information on TFIP11 limits the rationalisation of its biological role.
