Overexpression of histone deacetylase 8 (HDAC8) is associated with various diseases such as cancer. Thus, compounds that can modulate HDAC8 levels have therapeutic potential for these diseases. Based on the proteolysis targeting chimera (PROTAC) strategy, we designed and synthesized a series of HDAC8 degraders by tethering an HDAC6/8 dual inhibitor with pomalidomide (a cereblon ligand). Among them, compound ZQ-23 exhibited significant and selective degradation of HDAC8 with DC of 147 nM and D of 93%, and exhibited no effects on HDAC1 and HDAC3. Interestingly, we found that the degradation of target protein started at ∼2 h after treatment with ZQ-23 and the maximal degradation effect was achieved at 10 h. The HDAC8 level was partially recovered within 24 h. In addition, ZQ-23 had no degrading effects on HDAC1 and HDAC3 at all concentrations, but could dose-dependently increase the levels of acetylated SMC-3 (HDAC8 substrate). Mechanism study demonstrated that ZQ-23 degraded HDAC8 through the ubiquitin-protease pathway, rather than lysosome system. Collectively, these results suggest that ZQ-23 represents a novel PROTAC-based HDAC8 degrader worthy of further investigation.

The rheumatoid factor (RF) is a representative autoantibody against the crystallizable fragment (Fc) of denatured immunoglobulin (Ig) G that are primarily detected in patients with rheumatoid arthritis (RA). Although five types of tumor necrosis factor (TNF) inhibitors can be used to treat RA, no guidelines are available for selecting the appropriate inhibitor for treatment. High serum RF levels are associated with high disease activity, progressive joint destruction, life prognosis associated with organ damage, decreased treatment responsiveness to TNF inhibitors and other drugs, and low treatment retention rates.

However, existing methods for DNA methylation analysis are complex, time-consuming, and prone to DNA degradation. This work demonstrates selective capture of unmethylated DNAs using ZnO nanowires without chemical or biological modifications, thereby concentrating methylated DNA, particularly those with high methylation levels that can predict cancer risk.

Objective: It is hypothesized that the way nano- and micro-hybrid polymer-based composites are structured and cured impacts the way they respond to aging.
Material And Methods: A polymer-ceramic interpenetrating network composite (Vita Enamic/VE), an industrially polymerized (Brillinat CriosST/BC), and an in situ light-cured composite with discrete inorganic fillers (Admira Fusion5/AF5) were selected. Specimens (308) were either cut from CAD/CAM blocks (VE/BC) or condensed and cured in white polyoxymethylene molds (AF5) and subjected to four different aging conditions ( = 22): (a) 24 h storage in distilled water at 37 °C; (b) 24 h storage in distilled water at 37 °C followed by thermal cycling for 10,000 cycles 5/55 °C (TC); (c) TC followed by storage in a 75% ethanol-water solution; and (d) TC followed by a 3-week demineralization/remineralization cycling.
