If the antigen of interest is a cell-surface protein, flow cytometry or fluorescence-activated cell sorting (FACS) can be used to identify hybridomas secreting monoclonal antibodies to these proteins. Two alternative protocols are presented here-staining in individual tubes and staining in 96-well plates.

Microfluidic droplet sorting has emerged as a powerful technique for a broad spectrum of biomedical applications ranging from single cell analysis to high-throughput drug screening, biomarker detection and tissue engineering. However, the controlled and reliable retrieval of selected droplets for further off-chip analysis and processing is a significant challenge in droplet sorting, particularly in high-throughput applications with low expected hit rates. In this study, we present a microfluidic platform capable of sorting and dispensing individual droplets with minimal loss rates.

Structural variants of the synthetic opioid fentanyl are a major threat to public health. Following an investigation showing that many derivatives are poorly detected by commercial lateral flow and related assays, we created hapten conjugate vaccines using an immunogenic virus-like particle carrier and eight synthetic fentanyl derivatives designed to mimic the structural features of several of the more dangerous analogues. Immunization of mice elicited strong antihapten humoral responses, allowing the screening of hundreds of hapten-specific hybridomas for binding strength and specificity.
