There has been an explosion in scientific interest in using human-platelet-rich plasma (PRP) as a substitute of xenogeneic sera in cell-based therapies. However, there is a need to create standardization in this field. This systematic review is based on literature searches in PubMed and Web of Science databases until June 2021. Forty-one studies completed the selection criteria. The composition of PRP was completely reported in less than 30% of the studies. PRP has been used as PRP-derived supernatant or non-activated PRP. Two ranges could be identified for platelet concentration, the first between 0.14 × 10 and 0.80 × 10 platelets/µL and the second between 1.086 × 10 and 10 × 10 platelets/µL. Several studies have pooled PRP with a pool size varying from four to nine donors. The optimal dose for the PRP or PRP supernatant is 10%. PRP or PRP-derived supernatants a have positive effect on MSC colony number and size, cell proliferation, cell differentiation and genetic stability. The use of leukocyte-depleted PRP has been demonstrated to be a feasible alternative to xenogeneic sera. However, there is a need to improve the description of the PRP preparation methodology as well as its composition. Several items are identified and reported to create guidelines for future research.

Cell micro-patterning controls cell fate and function and has potential for generating therapeutically usable mesenchymal stromal cell (MSC) populations with precise functions. However, to date, the micro-patterning of human cells in a translational context has been impossible because only ruminant media supplements, e.g.

Human induced pluripotent stem cells (hiPSCs) can give rise to multiple lineages derived from three germ layers, endoderm, mesoderm and ectoderm. Definitive endoderm (DE) cell types and tissues have great potential for regenerative medicine applications. Current hiPSC differentiation protocols focus on the addition of soluble factors; however, extracellular matrix properties are known to also play a role in dictating cell fate.

Stem cell culture often requires various animal-derived components such as serum and collagen. This limits its practical use. Therefore, xeno-free (xenogeneic component-free) culture systems are receiving increased attention.

Article Synopsis
  • - The study explores the use of equine platelet lysate (ePL) as a replacement for fetal bovine serum (FBS) in growing muscle-derived mesenchymal stromal cells (mdMSCs) for regenerative medicine, addressing ethical and practical issues associated with FBS.
  • - Two innovative 2D and 3D culture models were tested, with the 3D model yielding a higher total cell count than the 2D model, while both models confirmed the multipotent capabilities of mdMSCs through differentiation assays.
  • - The mdMSCs showed promising immunomodulatory properties, significantly inhibiting myeloperoxidase (MPO) and reactive oxygen species (ROS) activity, especially in the

Article Synopsis
  • * A study was conducted using a rat model of type 2 diabetic nephropathy to evaluate the effects of mesenchymal stem cells (MSCs) and the angiotensin II receptor blocker irbesartan, both separately and in combination.
  • * The combined treatment of MSCs and irbesartan resulted in notable improvements in various kidney function parameters and reduced markers of kidney damage, suggesting that this combination could be a more effective therapy for DN than either treatment alone.
