An anionic water-soluble [2]biphenyl-extended-pillar[6]arenes modified with eight ammonium salt ions (AWBpP6) was successfully synthesized to establish a drug-drug conjugate supramolecular nanoprodrug (SNP) with a high drug-loading capacity. This SNP can generate a synergistic triple therapeutic effect of photodynamic therapy (PDT), photothermal therapy (PTT), and chemotherapy (CT; i.e., PDT-PTT-CT) with excellent biocompatibility.

Although antimicrobial peptides (AMPs) as a promising natural drugs can efficiently inhibit cervical cancer, poor bioavailability, low tumor selectivity, and non-selective toxicity still hinder its further application in vivo. In order to effectively address these challenges, we have developed a reactive oxygen species (ROS)-responsive targeting nanoprodrug designed for selective therapy of cervical cancer. Such nanoprodrugs (CEC-OxbCD) are fabricated by the supramolecular self-assembly of the modified β-cyclodextrin (β-CD) and AMPs.

A Supramolecular Self-Assembled Nanoprodrug for Enhanced Ferroptosis Therapy.

Ferroptosis can induce cell death that leverages Fe-triggered Fenton reactions within living organisms, leading to an excessive accumulation of lipid peroxides (LPOs) and inducing cell death. Ferroptosis can effectively circumvent the inevitable drug resistance encountered with traditional apoptotic therapies. However, several issues remain in the clinical application of ferroptosis anticancer therapy, primarily due to the poor efficiency of intracellular Fenton reaction.

Article Synopsis
  • Researchers developed a new type of nanoprodrug called BNN6@GBTC NPs, made from an amphiphilic prodrug (GBTC) and a nitric oxide donor (BNN6), which self-assemble into stable nanoparticles.
  • These nanoparticles can target HepG2 cancer cells specifically, thanks to galactose receptors on the cell surface, and generate reactive oxygen species (ROS) when exposed to light.
  • The combination of photodynamic therapy, chemotherapy through the release of camptothecin, and gas therapy using nitric oxide leads to a more effective cancer treatment while minimizing damage to healthy cells.

Fludarabine (FA) is still considered as a first-line chemotherapy drug for hematological tumors related to B lymphocytes. However, it is worth noting that the non-specific distribution and non-different cytotoxicity of FA may lead to irreversible consequences such as central nervous system damage such as blindness, coma, and even death. Therefore, it is very important to develop a system to targeting delivery FA.
