Injectable hydrogels exhibiting self-healing ability are promising carriers for controlled and sustained delivery in a minimally-invasive format for biomedical applications. We designed a polysaccharide-based double network hydrogel by mixing solutions of aldehyde-alginate (aAlg) and acrylic acid-chitosan (aCS) in the presence of adipic acid dihydrazide and FeCl that resulted in dual crosslinking mediated by Schiff base and ionic interactions. The hydrogel exhibited excellent thixotropic and self-healing properties with a high compressive fracture strength of ≈ 48 kPa. Encapsulated cells were viable within the hydrogel, and after their release from the degraded gel. The controlled release of Doxorubicin and Ciprofloxacin from the hydrogels established the gel as a delivery platform. The released drugs were effective in killing cancer cells or arresting the growth of both bacteria. This work presents a self-healing and injectable degradable hydrogel that may be used as a minimally-invasive platform for the delivery of drugs and cells.

Method: A total of 210 patients who underwent radial artery cerebral angiography at this center from September 2021 to May 2022 were selected, and their anatomical and clinical factors were analyzed to evaluate the correlation between these factors and the duration of radiographic exposure. Mutations in Leucine-rich repeat kinase 2 (LRRK2) and PTEN-induced kinase 1 (PINK1) are associated with familial Parkinson's disease (PD). LRRK2 phosphorylates Rab guanosine triphosphatase (GTPases) within the Switch II domain while PINK1 directly phosphorylates Parkin and ubiquitin (Ub) and indirectly induces phosphorylation of a subset of Rab GTPases. Herein we have crossed LRRK2 [R1441C] mutant knock-in mice with PINK1 knock-out (KO) mice and report that loss of PINK1 does not impact endogenous LRRK2-mediated Rab phosphorylation nor do we see significant effect of mutant LRRK2 on PINK1-mediated Rab and Ub phosphorylation.
