Background: Recent advancements in single-cell RNA sequencing have greatly expanded our knowledge of the heterogeneous nature of tissues. However, robust and accurate cell type annotation continues to be a major challenge, hindered by issues such as marker specificity, batch effects, and a lack of comprehensive spatial and interaction data. Traditional annotation methods often fail to adequately address the complexity of cellular interactions and gene regulatory networks.

Wound Healing in Older Adults After Major Lower Extremity Amputation.

Ann Vasc Surg

January 2025

Division of Vascular Surgery and Endovascular Therapy, Department of Surgery, Emory University School of Medicine; Surgical and Perioperative Care, Atlanta VA Healthcare System. Electronic address:

Introduction: The higher prevalence of multiple chronic conditions and frailty among older adults may increase the physiologic demand required for wound healing after a major lower extremity amputation (LEA). After below knee amputations (BKA), patients generally have improved prosthetic fitting rates, postoperative ambulation, and quality of life compared to an above knee amputation (AKA). However, the benefit of a BKA must be weighed against the risk of wound complications.

Purpose: The purpose of this study was to compare the surgical outcomes of anterior cruciate ligament (ACL) reconstruction using hamstrings autograft (HA) plus lateral extra-articular tenodesis (LET), isolated all-soft-tissue quadriceps autograft (QA), and isolated HA.

Methods: A retrospective review was performed comparing high risk patients undergoing ACL reconstruction with isolated HA, isolated QA, or HA+LET from August 2013 to January 2023. High risk patients, as determined by high grade pivot shift or generalized ligament laxity, with at least 2 years of follow up were included.

Large language models vs human for classifying clinical documents.

Int J Med Inform

January 2025

College of Science and Engineering, James Cook University, Townsville, 4811, QLD, Australia. Electronic address:

Background: Accurate classification of medical records is crucial for clinical documentation, particularly when using the 10th revision of the International Classification of Diseases (ICD-10) coding system. The use of machine learning algorithms and Systematized Nomenclature of Medicine (SNOMED) mapping has shown promise in performing these classifications. However, challenges remain, particularly in reducing false negatives, where certain diagnoses are not correctly identified by either approach.

Background: The internet is a key source of health information, but the quality of content from popular search engines varies, posing challenges for users-especially those with low health or digital health literacy. To address this, the "tala-med" search engine was developed in 2020 to provide access to high-quality, evidence-based content. It prioritizes German health websites based on trustworthiness, recency, user-friendliness, and comprehensibility, offering category-based filters while ensuring privacy by avoiding data collection and advertisements.
